2种人卵巢癌裸鼠皮下移植瘤模型制备方法的比较

【摘要】 目的比较直接接种人卵巢癌COC1细胞和COC1细胞经体内传代后接种Balb/c-nu裸鼠皮下移植瘤模型制备方法的成瘤率。方法雌性突变系Balb/c-nu小鼠40只,分为对照组及实验组,每组20只。COC1细胞注射于裸鼠一侧背部皮下,对照组细胞培养后即接种,观察统计数据后处死,取瘤体制成单细胞悬液再接种于实验组裸鼠,比较两种不同方法接种的成瘤率,并观察成瘤时间、瘤重及肿瘤生长速度。结果实验组成瘤率为70%,对照组仅为30%。结论经过裸鼠体内传代后的COC1细胞用于制备人卵巢癌裸鼠皮下移植瘤模型具有较高的成瘤率。

【Abstract】 Objective To establish the best way of tumor-bearing Balb/c-nu nude mice model with Epithelial varian adenocarcinoma cell line COC1.Methods Balb/c-nu nude mice were divided into experimental group(EG) and control group(CG);COC1(suspended cell)cells were injected into nude mice subcutaneous layers of back.In EG,COC1 cells were passaged in vivo of nude mice,but COC1 cells in CG were not passaged.The rate of tumor formation of different methods was compared and the time of tumor formation,the weight annd growth speed of tumor were observed.Results The rate of tumor formation were 70% in EG and 30 % in CG.Conclusion Through using the passaged COC1 cells,the rate of tumor formation was highly improved.
