低温-复温对大鼠肾脏细胞凋亡的影响及丹参的保护作用

【摘要】 目的探讨低温-复温过程对大鼠肾脏细胞凋亡的影响及丹参的保护作用。方法建立Wistar大鼠肾脏离体非循环灌注模型,随机分为4组(n=12):对照组、低温保存组、低温-复温组、丹参实验组。采用TUNEL法检测肾组织细胞凋亡情况,免疫组化SABC法观察Caspase-3、Bcl-2蛋白表达情况。结果低温-复温组细胞凋亡指数为(53.2±17.1)%,显著高于对照组(0.2±0.1)%和低温组(0.3±0.1)%(P<0.01);丹参实验组细胞凋亡指数(36.7±14.6)%明显低于低温-复温组(53.2±17.1)%(P<0.05);丹参实验组Caspase-3表达(5.4±1.3)%低于低温-复温组(11.9±6.2)%(P<0.05),而Bcl-2表达(1.8±0.3)%高于低温-复温组(0.7±1.5)%)(P<0.05)。结论低温-复温过程可造成肾脏细胞凋亡显著增加;丹参可减少肾脏复温相关性细胞凋亡,其机制可能与抑制Caspase-3活性、上调Bcl-2基因表达有关。

【Abstract】 Objective To investigate protective effect of Salvia miltiorrhiza on rewarming-associated apoptosis of rat kidney.Methods Isolated nonreciuculated perfusion model of rat kidney was established.Forty-eight Wistar rats were randomly devided into 4 groups(n=12):control group,cold preservation group,cooling/rewarming preservation,and salvia miltiorrhiza group.The apoptosis of kidney tissue was examined with TUNEL assay,and the expressions of Caspase-3 and Bcl-2 were detected with immunochemical method.Results The apoptosis index(AI) of kidney was(53.2±17.1)% in cooling/rewarming group,which was higher than that in normal group(0.2±0.1)%(P<0.01) and in cold preservation group(0.3%±0.1%)(P<0.01).But it was much lower in Salvia miltiorrhiza group(36.7±14.6)%(P<0.05).Compared with cooling/rewarming group(11.9±6.2)% vs(0.7±1.5)%,the protein expression of Caspase-3 in Salvia miltiorrhiza group(5.4±1.3)%(P<0.05) was decreased,while that of Bcl-2 group(1.8±0.3)%(P<0.05) was increased remarkably.Conclusion The process of cooling/rewarming may accelerate the apoptosis of kidney,which could be ameliorated by Salvia miltiorrhiza through inhibiting Caspase-3 activity and upregulating Bcl-2 expression.
