生芪颗粒对胰岛素抵抗糖尿病大鼠AT1、Ang Ⅱ、ET-1的影响

【摘要】 目的:观察生芪颗粒干预胰岛素抵抗大鼠血清ET-1、AngⅡ和心脏AT1等指标水平变化,探讨中药对胰岛素抵抗血管内皮功能的影响机制。方法:选用健康雄性Wistar大鼠60只,按随机数字表法分为6组:生芪颗粒低、中、高剂量组、罗格列酮组、正常组和模型组。正常组和模型组给予枸椽酸钠溶液。生芪颗粒低、中、高剂量组以人鼠单位体重用药量直接换算后剂量的5,10,20倍灌胃生芪颗粒溶液。罗格列酮组:灌胃100 g/L的马来酸罗格列酮悬浊液。测定治疗前后血清ET-1、AngⅡ变化;免疫组化法测定治疗前后心脏AT1的变化。结果:生芪颗粒高剂量组、罗格列酮组血清ET-1、AngⅡ与正常组比较均无显著差异(P>0.05),生芪颗粒中、高剂量组、罗格列酮组血清ET-1、AngⅡ与模型组存在显著差异(P<0.05);生芪颗粒各组、罗格列酮组、模型组心脏AT1和正常组比较均有显著性差异(P<0.05),与模型组相比,生芪颗粒中、高剂量组和罗格列酮组均有明显的下降(P<0.05)。结论:生芪颗粒可降低胰岛素抵抗大鼠血清ET-1、AngⅡ和心脏AT1的免疫组化表达,作用效果与罗格列酮相当,并呈剂量依赖性。

【Abstract】 To observe the effects of Shengqi Granule on AT1,Ang and ET-1 of insulin resistant rats with diabetes,and to probe the influence of Chinese drugs on insulin resistance against blood endothelial function.Methods: Sixty healthy male Wistar rats were randomly divided into 6 groups: lowdosage,middle-dosage and high-dosage of Shengqi Granule group,Rosiglitazone group,normal group and model group.The normal group and model group were given Sodium Citrate Solution.Shengqi Granule group were given gastric perfusion of Shengqi Granule solution.Rosiglitazone group were given gastric perfusion of Rosiglitazone maleate Suspension.The change of serum ET-1 and AngⅡ were examined before and after the treatment.The immunohistochemical therapy was used to examine the change of heart AT1 before and after the treatment.Results: There existed no significant difference(P>0.05) for the serum ET-1 and AngⅡ of high-dosage of Shengqi Granule group and Rosiglitazone group compared with that of the normal group.There existed a significant difference(P<0.05) between the serum ET-1 and AngⅡ of middle-dosage and high-dosage of Shengqi Granule groups and those of Rosiglitazone group.There existed a significant difference(P<0.05) between the heart AT1of Shengqi Granule groups,Rosiglitazone group and the model group and that of the normal group.Conclusion: Shengqi Granule can decrease the immunohistochemical expression of serum ET-1 and AngⅡ of IR rats and heart AT1,and its therapeutic effects are equivalent with that of Rosiglitazone group and there exists a dosage-dependent relationship.
