甲硝唑体外抗阴道毛滴虫的电镜观察

【摘要】 目的:观察甲硝唑对阴道毛滴虫超微结构的影响。方法:在2.5×106个.mL-1阴道毛滴虫培养液中加入甲硝唑(5mg/L),37℃培养3~5h,利用扫描和透射电镜观察甲硝唑对阴道毛滴虫超微结构的影响。结果:甲硝唑作用于阴道毛滴虫后,对其细胞膜及氢化酶体影响不明显;阴道毛滴虫细胞质出现大量空泡、裂隙,内质网消失、细胞核变形及自噬空泡,最终虫体变性坏死。结论:甲硝唑对阴道毛滴虫细胞内结构的破坏是导致虫体死亡的主要原因。

【Abstract】 Aim:To observe ultrastructural influence of metronidazole on Trichomonas vaginalis(T.vaginalis) in vitro.Methods:5 mg/L metronidazole was added into liver extract solution medium that contained 2.5×106 parasites/mL.The trophozoites of T.vaginalis were cultivated at 37 ℃ for 3~5 hours.The parasites treated with this drug were observed by scanning and transmission electron microscope.Results:The surface of T.vaginalis trophozoites treated with the drug only showed many small bubbles.But the cytoplasms of the organisms were damaged seriously.Many vacuoles,crevices,arear of no structure and autophagous vacuole were visible in the cytoplasms of the parasites.Meanwhile endoplasmic reticulums were disorganized and disappered.The ring space was formed around the nucleus.The nuclear membrane was damaged.The morphology of the nucleus was changed.The organisms treated with the drug lost the phagocytic ability.Finally the parasites were denatured and died.Conclusion:Metronidazole destroyed the internal structures of T.vaginalis trophozoits that was the main source of the death.The effect of metronidazole on the cellular membrane and hydrogenosomes of the parasites was not obviously.
