钩吻B2组分对Hela细胞增殖和细胞周期的影响

【摘要】 目的:探讨钩吻醇提-氯仿萃取(GW-B2)组分对Hela细胞生长与增殖以及细胞周期的影响。方法:采用XTT法分析细胞生长与增殖,Timelapse显微镜观察细胞生长和形态,流式细胞仪检测凋亡和细胞周期,综合评价GW-B2组分抗Hela细胞的作用。结果:GW-B2具有明显抑制Hela细胞生长与增殖的作用,1μL/mL GW-B2组分处理的Hela细胞生长速度明显降低,在2~5μL/mL的剂量范围内,细胞生长与增殖能力基本完全被抑制。GW-B2组分处理后,Hela细胞主要的形态学变化为贴壁能力丧失,悬浮细胞增多,细胞分裂阻滞,部分细胞崩解破碎。GW-B2组分能明显的诱导Hela细胞凋亡,1μL/mL、2μL/mL和5μL/mL GW-B2组分处理72h后,Hela细胞的凋亡率分别可达8.16%、36.83和61.32%,而对细胞周期的影响主要是G2/M期阻滞,同时对G0/G1期也有一定的阻滞作用。结论:GW-B2组分具有抑制Hela细胞增殖、诱导凋亡,具有明显的抗肿瘤细胞的作用;GW-B2组分对细胞周期的影响主要是G2/M期阻滞,随后可诱导细胞的凋亡。

【Abstract】 Objective To study on effect of compound of ethanol and chloroform extraction from Gelsemium elegans Benth(GW-B2 component) on Hela cells proliferation and cell cycle.Methods Cell proliferation was detected by XTT method,cell growth morphological changes was observed with Time-lapse inversion microscope,apoptosis and cell cycle was analyzed by flow cytometer,comprehensive evaluated the effects of extract of Gelsemiumon cell growth and proliferation.Results GW-B2 component obviously inhibited Hela cell growth and proliferation,growth speed of Hela cell was decreased obviously at concentration 1μL/mL,and cell growth and proliferation ability was inhibited almost completely at concentration 2μL/m or 5μL/mL.Distinct morphological abnormity can be observed duo to treatment with GW-B2 component,such as adhesive capability of cells lose,suspension cells increase,cell division blockage,and a part of cells broken and fragmentized.Apoptosis can be distinctly induced by GW-B2 component,apoptosis rate were respectively 8.16%,36.83 and 61.32% when Hela cells was treated with GW-B2 component at concentration 1μL/mL,2μL/mL and 5μL/mL for 72h.Primary effect of GW-B2 component on cell cycle was G2/M stage blockage,and definite G0/G1 stage blockage can be also observed at same time.Conclusion GW-B2 component can inhibited cell proliferation,induced cell apoptosis,which indicated its unambiguous effects on tumor cell.Primary effects on cell cycle of GW-B2 component was G2/M stage retardation,and then apoptosis can induced.
