【摘要】 目的:探讨碱性成纤维细胞生长因子(basic fibroblast growth factor,FGF-2)对小细胞肺癌(small cell lung cancer,SCLC)细胞株NCI-H446中survivin的表达及Smac亚细胞定位的影响。方法:West-ern印迹分析FGF-2对NCI-H446中凋亡抑制蛋白survivin表达的影响。Western印迹和免疫荧光分析FGF-2对线粒体释放的促凋亡蛋白Smac的亚细胞定位的影响。流式细胞术检测NCI-H446的细胞凋亡百分率。Hoechst33258染色观察FGF-2对NCI-H446细胞凋亡的影响。结果:FGF-2能明显诱导NCI-H446细胞中survivin蛋白的表达,survivin的表达水平与FGF-2作用的浓度和时间明显相关。血清饥饿诱导的Smac蛋白从线粒体的释放可被FGF-2所抑制。FGF-2能明显抑制血清饥饿诱导的凋亡。结论:FGF-2可显著提高SCLC细胞株NCI-H446中凋亡抑制蛋白survivin的表达水平,并显著抑制血清饥饿诱导的促凋亡蛋白Smac释放入胞浆。这可能与其抑制肿瘤细胞凋亡有关。更多还原

【Abstract】 Objective To investigate the effect of basic fibroblast growth factor(FGF-2)on survivin and subcellular location of Smac in human small cell lung cancer(SCLC)cell NCI-H446.Methods Western blot was used to detect the expression of survivin protein induced by FGF-2.The release of Smac from mitochondria to cytoplasm affected by FGF-2 was observed by Western blot and immunofluorescence.Apoptosis of NCI-H446 cells was detected with flow cytometry and Hoechst 33258 staining.Results The expression of survivin could be up-regulated in response to FGF-2 treatment in NCI-H446 cells,and the level of survivin expression is related to the concentration and time of FGF-2 treatment.FGF-2 could inhibit the release of Smac from the mitochondria to cytoplasm induced by serum starving.FGF-2 could inhibit the apoptosis induced by serum starving.Conclusion FGF-2 up-regulates the expression of survivin protein in NCI-H446 cells,and blocks the release of Smac from mitochondria cytoplasm.Survivin and Smac might play important roles in the apoptosis inhibited by FGF-2.更多还原
