脑膜瘤MMP-29、及TIMP-1 mRNA表达与瘤周水肿的相关性研究

【摘要】 目的研究脑膜瘤中基质金属蛋白酶2、9(MMP-2、9)及其抑制因子TIMP-1mRNA表达与其瘤周水肿(PTE)的相关性。方法36例经MRI检查及手术、病理证实的脑膜瘤,根据PTE程度分为2组:无或轻度水肿组和中重度水肿组,将手术标本以免疫组化染色和原位杂交方法对MMP-2、9和TIMP-1 mRNA表达行半定量检测,分析与脑膜瘤瘤周水肿之间的相关性。结果脑膜瘤的瘤周水肿程度与MMP2(P<0.01)和MMP-9(P<0.005)的表达强度差异有统计学意义。而TIMP-1 mRNA的表达在无或轻度水肿组和中重度水肿组之间无显著性差异(P>0.05)。结论MMP-2、9的表达及MMP/TIMP比例失衡在脑膜瘤的高侵袭性和瘤周水肿程度的发生中起关键作用,PTE作为脑膜瘤MRI影像的一个重要特征,因反映了肿瘤的侵袭程度,故可作为临床预后评价的指标之一。

【Abstract】 Objective To determine whether the expressions of matrix metalloproteinases-2,9(MMP-2,9) and tissue inhibitor of metalloproteinase-1(TIMP-1) mRNA were correlated with peritumoral edema(PTE) in human meningioma.Methods Thirty-six meningioma specimens were collected for retrospective analysis,which were testified by MRI and proved by operation and pathology.According to the degrees of PTE,the patients were divided 2 groups: one with minimum to mild edema and the other with moderate to mild edema.Protein expressions of MMP-2,9 and mRNA expression of TIMP-1 were semi-quantified in tissue sections by immunohistochemical techniques and in situ hybridization,the results of staining were compared with the degrees of PTE.Results A statistical significance was apparent between the expressions of MMP-2(P<0.05),MMP-9(P<0.05)and peritumor edema(PTE) of meningioma,and there was no statistical significance between the expression of TIMP-1 mRNA and the degree of PTE.Conclusion MMP-2,9 as well as MMPs/TIMP ratio may play an important role in the invasiveness of meningioma and PTE degree.PTE may predict the prognosis of meningiomas.
