大鼠骨髓基质细胞移植治疗胶质瘤的实验研究

【摘要】 目的将Wistar大鼠骨髓基质细胞体外移植入C6胶质瘤大鼠模型,观察其生物学特性及大鼠存活状态。方法建立C6载瘤模型和骨髓基质细胞移植治疗模型;待术后生长至3w和4w时行MRI检测;不同时间段内大鼠脑标本行免疫组化染色。结果骨髓基质细胞移植组模型平均生存时间为4.03w;对照组平均生存时间为3.88w;且其平均肿瘤体积与对照组无明显差异。大鼠体内移植体外培养5月余的骨髓基质细胞未发现肿物生成。骨髓基质细胞包绕在胶质瘤周围,对胶质瘤细胞有定向靶向作用。结论骨髓基质细胞对胶质瘤有定向靶向作用,可以安全的作为生物载体转染目的基因治疗胶质瘤。

【Abstract】 Objective To observe the biology character and the state of life by Wistar rat bone marrow stromal cells transplantation into the models of C6 glioma.Methods We established 25 models of C6 glioma and 35 models of bone marrow stromal cells transplantation. 5 rats were selected randomly from above two model groups for magnetic resonance imaging at 3 weeks and 4 weeks after established the glioma models. We got the brain tissue stained HE and BrdU at different times. Results Mean survival time in the neural stem cells transplanted group was 4.03w which was a little longer than that in control group (3.88w)and the mean tumor volume was the same as the control group. There was no tumour growth in the Wistar rat after transplanted the bone marrow stromal cells cultured in vitro more than 5 months.There were 5-bromodeoxyurid -ine-positive bone marrow stromal cells arround the the glioma and it displayed tropism for the glioma. Conclusion It is safe to transplant the rat bone marrow stromal cells for the homogeneity rats. The bone marrow stromal cells display tropism for the glioma.They may be used to transfect the gene to cure the glioma.
