【摘要】 为了弄清我国不同地区猪繁殖与呼吸综合征病毒(PRRSV)的来源及其遗传学背景,为疫苗学研究提供理论根据,在获得CH-1a株完整的基因组以后,从流行于我国山东(SD)和黑龙江大庆(DQ)地区疑似PRRS的猪体内分离到PRRSV,在Marc-145细胞上盲传5~6代,细胞出现明显病变以后收获病毒液,提纯并提取全病毒RNA,经过反转录、PCR扩增获得结构基因ORF 2~6的目的基因片段,然后与pMD-T载体连接,转化,得到阳性质粒后进行测序,并将其与CH-1a株进行比较分析,同时对这两个毒株的结构基因组的理化性质进行分析。结果表明:流行于我国山东和大庆地区的PRRSV结构基因ORF 2~6与CH-1a株核苷酸的同源性均在94%~99%之间,氨基酸的同源性也在90%以上,但从DQ和SD株结构基因编码的蛋白来看,这两个毒株与CH-1a株的结构基因所编码的蛋白在氨基酸组成以及等电点方面还有一定的差异,表明流行于我国的PRRSV毒株均属于美洲型。

【Abstract】 To source and genetic background of porcine reproductive and respiratory syndrome virus(PRRSV) from different parts of China were clarified.Two strains of PRRSV were isolated from the swine infected with PRRSV in Shangdong Province and Daqing District.The PRRSV was cultured on Marc-145 cells for 5~6 passages,then harvested and purified.The whole virus RNA was extracted.The target genes fragments ORF 2~6 were acquired by PCR and then cloned into the pMD-18T vector.The positive recombinant plasmids were acquired after being transformed.The physical and chemical properties of the structural gnome were analyzed.The structural genome of prevalent PRRSV in Shandong Province and Daqing District were 94%~99% identity with those of CH-1a strain,the identities of amino acid were also beyond 90%.However,the proteins encoding these structural genes show some difference,especially in the composition of amino acid.PRRSV prevalent in China belongs to those of VR-2332.
