重组蛋白对工程菌的保护作用

【摘要】 为考察重组蛋白对强氧化剂环境中工程菌的保护作用,分别在含百草枯及维生素K3(VitK3)的强氧化剂和不含强氧化剂的培养基中培养重组蛹虫草超氧化物歧化酶菌株和对照菌株并诱导表达,测定菌体密度(A600)及菌体内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的酶活变化。结果表明:重组菌株对百草枯和VitK3引起的生长抑制作用具有抗性,百草枯和VitK3对重组蛋白的表达没有影响,重组菌株的A600、SOD酶活力明显高于对照菌株,CAT酶活力略高于对照菌株。说明外源基因在E.coli中的高效表达提高了菌体抵抗自由基引起的损伤的能力。

【Abstract】 In order to investigate the protective role of the expression of recombinant protein in engineering strain against oxidative damage,recombinant cm-SOD strain and control strain were cultivated in the medium with or without paraquat and vitamin K3(VitK3).Cell density A600,superoxide dismutase(SOD) activity and catalase(CAT) activity were measured.Results showed that the recombinant strain could build up resistance to paraquat and VitK3-mediated inhibition of growth.The A600 of recombinant strain was significantly higher than that of control strain.The expression of recombinant protein was not influenced by paraquat or VitK3.Thus,SOD activity of recombinant strain was significantly higher than that of control strain,and its CAT activity was a bit higher than that of control strain when cultured in the same condition.Collectively,these data suggest that the overexpression of heterologous source gene in E.coli provides protection against damage attributable to free radicals.
