超高压处理对大肠杆菌DH5α质粒DNA的影响
Effect of UHP processing on the plasmid DNA of the Escherichia coli DH5α
【摘要】 文章研究了超高压处理对大肠杆菌DH5α菌株质粒pMD-18DNA的影响。大肠杆菌DH5α在不同的压力下于25℃处理15 min后,提取其质粒pMD-18DNA,检测质粒在260 nm和280 nm处的吸光度得到其纯度,并进行了琼脂糖凝胶电泳。实验发现:大肠杆菌DH5α质粒pMD-18DNA经100 MPa、200 MPa处理后其A260、A280与未经高压处理样相比吸光度下降;而经300 MPa4、00 MPa和500 MPa处理后其吸光度与未经高压处理样相比吸光度上升。通过琼脂糖凝胶电泳分析,大肠杆菌DH5α质粒DNA经高压(300 MPa、400 MPa和500 MPa)后条带增多。实验结果表明超高压处理影响了质粒DNA的结构。
【Abstract】 The paper investigates the effect of high hydrostatic pressure treatment(100 MPa,200 MPa,300 MPa,400 MPa and 500 MPa) under the temperature of 25℃ with the dwell time of 15 min on the plasmid pMD-18 DNA extracting from Escherichia coli DH5α.The plasmid was studied by detecting its UV absorbency and purity in 260 nm and 280 nm and by DNA agarose gel electrophoresis.The results are as follows: the absorbency of the plasmid pMD-18 DNA in 260 nm and 280 nm treated by 0.1 MPa is larger than that treated by 100 MPa and 200 MPa but less than that treated by 300 MPa,400 MPa and 500 MPa under the treatment temperature 25℃ and dwell time 15 min;and by DNA agarose gel electrophoresis,electrophoresis bands of the Escherichia coli DH5α plasmid pMD-18 DNA are increased after being treated by high hydrostatic pressure(300MPa,400MPa and 500MPa).The results mean that the configuration of the Escherichia coli DH5α plasmid DNA is changed.
