锌指结构9、结缔组织生长因子在大鼠肝星状细胞中的表达
Expression of ZF9 and CTGF in Primary Hepatic Stellate Cells of Rat Liver Fibrosis
【摘要】 目的:通过研究肝纤维化大鼠原代肝星状细胞锌指结构9(ZF9)、结缔组织生长因子(CTGF)的表达及其动态变化,初步探讨CTGF、ZF9在肝星状细胞活化中的作用。方法:32只雄性SD大鼠皮下注射40%植物油CCl4,每周2次,并分别于注射1,4,8周后作大鼠肝原位灌注,分离肝星状细胞,Western blot法检测CTGF蛋白,RT-PCR检测ZF9 mRNA水平。结果:正常大鼠肝星状细胞ZF9基因可见弱表达,注射CCl41周后其表达增强,注射CCl44周后转录明显增强,8周时仍高表达。正常大鼠肝星状细胞CTGF蛋白无明显表达,模型组大鼠皮下注射CCl41周后,蛋白出现少量表达,注射CCl44周后,表达进一步增加,第8周CTGF蛋白表达更强。结论:ZF9、CT-GF基因随着肝损伤的加重,在肝星状细胞中的表达逐渐增加,共同促进肝纤维化的发生。
【Abstract】 Objective: To investigate the role of ZF9 and CTGF in activation of hepatic stellate cells in liver fibrosis rats.Methods: Rat liver fibrosis model was made by subcutaneous injection of CCl4.Rat hepatic stellate cells were isolated by in situ portal vein perfusion and density gradient centrifugation after 1,4 and 8 weeks of injection of CCl4.Western blot was carried out to detect the expression of CTGF.The mRNA level of ZF9 was evaluated by RT-PCR.Results: No expression of CTGF was detected in hepatic stellate cells of normal rats.There was mild expression after one week of subcutaneous injection of CCl4.It was stronger at the 4th week and strongest at the 8 th week after injection of CCl4.ZF9 gene express was mild in normal rat liver and in model group at the first week after injection of CCl4.Its expression became stronger at the 4th and 8th week.Conclusion: Expression of ZF9 and CTGF became stronger in hepatic stellate cells of rats with the development of liver injury and play important roles in activation of hepatic stellate cells.
