肌动蛋白基因在棉花纤维中的作用研究
Transformation of GhACT1 RNAi and Its Effect on Fiber Length of Cotton(Gossypium hirsutum)
【摘要】 运用农杆菌介导法将肌动蛋白RNAi表达载体PBI121-GhACT1转化棉花,经胚胎发生获得再生植株。非转化组培再生苗作为对照。以NPTII制备探针,Southern杂交结果表明,获得8个独立遗传转化系,其中3个系有2个拷贝插入;其余6个系为多拷贝。显微镜下观察开花后第5天的幼胚珠孔端,对照纤维长度约是RNAi载体转基因的3~5倍。成熟后绒长F测验结果表明,RNAi载体转化株与对照差异显著,RNAi转基因植株的绒长显著短于对照。说明GhACT1基因对纤维有延缓和抑制作用,并且主要作用于纤维发育早期。
【Abstract】 Introduced the actin RNAi(interference) plasmid PBI121-GhACT1 into Coker 312 by Agrobacterium mediate method,and abtained the regenerants through somatic embryogenesis.NPTII as probes to hybridize,Southern result showed that there were two copies inserted into A5,A6 and A14 transgenic lines,the other six lines had multiple copies insertion.That declared these lines were transgenic positive lines.Observed under microscope,the young fiber on RNAi transgenic seeds was 3-5 fold shorter than that on tissue culture immature seeds.Analysis mature fiber length by statistic F test,result showed that F value beyond F0.05=4.10,the fiber length of RNAi transgenic lines was significant shorter than tissue cultural fiber.This declared that the GhACT1 gene played a significant role in fiber length,and function mainly at early elongation period.
