骨髓间充质干细胞复合去抗原牛松质骨修复兔桡骨骨缺损
Effects of Mesenchymal Stem Cells and Antigen-extracted Bovine Cancellous Bone Complex on Repairing the Radial Defect in Rabbits
【摘要】 目的:研究复方接骨中药含药血清培养诱导的骨髓间充质干细胞复合去抗原牛松质骨载体修复兔桡骨骨缺损的作用机制。方法:取4月龄大耳白兔24只随机分为A、B2组,将A、B组每只兔一侧前肢制成桡骨骨缺损模型,A组于骨缺损处植入复方接骨中药含药血清培养诱导的骨髓间充质干细胞与去抗原牛松质骨复合体;B组于骨缺损处植入单纯去抗原牛松质骨。术后3、6、12周分别取材,将术侧桡骨完整取出,进行大体观察、X线片及组织学检查,麦胚凝集沉淀法测定骨源性碱性磷酸酶活性。结果:大体观察及X线片显示:A组骨痂生长良好,骨缺损修复完成;B组骨缺损部分修复,部分为骨不连接。组织学检查:A组新生骨量较多,见大量成骨细胞,骨缺损与断端形成骨性连接。B组骨小梁间隙可见纤维组织长入,较多软骨团出现,编织骨数量多。统计学分析表明骨源性碱性磷酸酶活性A组显著高于B组(P<0.01)。结论:复方接骨中药含药血清培养诱导的骨髓间充质干细胞复合去抗原牛松质骨载体能够较好地修复兔桡骨骨缺损。
【Abstract】 Objective:To explore the effects of mesenchymal stem cell(MSC)and antigen-extracted bovine cancellous bone(BCB) complex cultured and induced by serum containing Traditional Chinese Compound Bone Healing Recipe(TCCBHR)on repairing the radial defect in rabbits.Methods:24 four-month-old rabbits were divided into Group A and B.In Group A,the MSC-BCB was implanted in the place of radial defect in unilateral forearm.In Group B,the BCB was implanted in the place of radial defect in unilateral forearm only.Then the radial was taken from Group A and B respectively 3,6 and 12 weeks after operation.The normal morphologic observation,X-ray and histologic checks were taken and the activity of BALP was determined by wheat=embryo curdling and deposition method.Results:In Group A,the normal morphologic observation and X-ray manifestation showed that the callus grew well and bone defect repair was completed,but in Group B,bone defect repair was completed,but some with nonunion.The histologic check showed that in Group A,there were a great number of osteoblast and new bone mass,bone union appeared in the place of bone defect.In Group A,the fibrous tissue grew well at the place of bone grider clearance.There was a great number of cartilage gland alveolus and woven bones.The statistic analysis showed that the bone alkaline phosphatase(BALP)activity in Group A was obviously greater than that of the B group(P<0.01).Conclusion:The MSC-BCB complex cultured and induced by serum containing Traditional Chinese Compound Bone Healing Recipe(TCCBHR)can repair radial defect in rabbits.
