【摘要】 目的:观察骨宝含药血清对SD大鼠成骨细胞(OB)中护骨素(OPG)mRNA表达的影响。方法:①将20只10月龄SD雌性大鼠随机分为2组,每组10只,分别为骨宝组和生理盐水对照组,制备含药血清和空白血清,MTT法确定最佳添加剂量;②取1d龄SD大鼠颅骨分离培养OB,分为骨宝含药血清组和对照组,磷酸对硝基苯酯法(PNPP法)检测OB碱性磷酸酶(ALP)活性,荧光定量聚合酶链式反应(FQ-PCR)检测OB中OPG mRNA的表达。结果:①15%浓度的骨宝含药血清对OB的增殖具有明显的促进作用(P<0.05);②骨宝含药血清组中OB的ALP活性2.527±0.185高于对照组1.839±0.040(P<0.05),骨宝含药血清组中OB的OPG mRNA表达量1.457±0.031高于对照组0.852±0.090(P<0.05)。结论:骨宝含药血清能上调OB中OPGmRNA的表达,增强OB的ALP活性,可能是骨宝防治骨质疏松症的作用机制之一。更多还原

【Abstract】 Objective:To explore the effects of Gubao-containing serum on ALP activity and OPG mRNA expression in osteoblasts(OB).Methods:① Twenty female SD rats at 10 months of age were divided into two groups to prepare Gubao-containing serum and control serum.The optimal dose of Gubao was detected using methyl thiazolyl tetrazolium(MTT)assay.② SD rats at 1 day of age were divided into Gubao group and control group,and cranial bones of these rats were used to isolate OBs for culture.The ALP activity of OB was detected by p-nitrophenylphosphate(PNPP),and OPG mRNA expression of OB was detected by fluorescent quantitative polymerase chain reaction(FQ-PCR).Results:①Serum containing 15% Gubao showed significant stimulation for the growth of OBs(P<0.05).② The ALP activity and OPG mRNA expression were frankly enhanced in Gubao group compared with control group(2.527±0.185 vs 1.839±0.040,1.457±0.031 vs 0.852±0.090,respectively,both P<0.05).Conclusion:The up-regulated OPGmRNA expression and enhanced ALP activity in Gubao-containing serum may be part of mechanisms underlying the anti-osteoporotic effects of Gubao.更多还原
