羟自由基降解法对海带硫酸多糖生理活性的影响

【摘要】 目的:研究羟自由基降解对海带硫酸多糖理化性质和生理活性的影响。方法:分别利用Harber-Weiss和Fenton体系降解海带硫酸多糖。采用硫酸酚法、比浊法、HPLC法研究降解前后多糖的糖含量、硫酸根含量、分子量等理化性质的变化。采用促小鼠脾淋巴细胞增殖实验研究降解前后多糖的生物学功能变化。结果:经羟自由基降解,海带硫酸多糖的分子量明显降低,降解前后多糖的各项理化性质变化不大。降解前后多糖都能明显促进小鼠脾淋巴细胞增殖,两者作用效果差异不明显(P>0.05),但降解后多糖能更快的发挥作用。结论:羟自由基降解对海带硫酸多糖的理化性质无影响,但能更好的发挥海带硫酸多糖的生理功能。

【Abstract】 Objective: To explore the impacts of hydroxyl radical degradation on physicochemical features and biological activities of Laminaria polysaccharide sulfate (LPS). Methods: The Harber-Weiss and Fenton systems were used for hydroxyl radical degradation of LPS. Molecular weight, ratio contents of saccharide and sulphates (SO4 2-) were assessed with turbidimetry, phenol-sulphuric acid method and high performance liquid chromatography. The biological activities of LPS before and after degradation were evaluated through splenic T lymphocyte proliferative responses in mice. Results:After degradation, the LPS showed significantly lower molecular weight but no remarkable changes in physicochemical features. Degraded LPS consistently promoted splenic lymphocyte proliferation in mice, with no difference in biological activities compared with before degradation, but that degradation caused shorter time-to-effect for LPS. Conclusion: Hydroxyl radical degradation has no impacts on physicochemical features but causes a shorter time to biological effect of LPS.
