应用RAPD技术探讨苦丁茶冬青种质材料的起源
A STUDY ON THE ORIGIN OF GERMPLASM MATERIALS OF ILEX KUDINGCHA C.J.TSENG BASED ON RAPD ANALYSIS
【摘要】 以15份原产地明确的苦丁茶冬青野生种质材料作为参照,利用RAPD-PCR技术对26份苦丁茶冬青栽培材料的原产地进行了分析.从40个10 bp引物中筛选出16个多态性良好的引物,用该16个引物对上述41份苦丁茶冬青种质材料进行PCR扩增,共得到238条DNA谱带,其中多态性带190条,占扩增出的总带数之79.8%.根据扩增结果计算了各份材料之间的Jaccard遗传相似系数,并用UPGMA法构建了聚类树状图.供试的41份种质材料在聚类图中明显地分为两个类群,即海南岛类群和广西南宁类群.26份原产地待查的种质材料有12份归聚于海南岛类群,说明它们的起源地在海南岛境内;另14份原产地待查的种质材料分布归聚于广西南宁类群,说明它们的原产地在广西南宁地区或毗邻区域内.所有供试的41份苦丁茶冬青材料其遗传相似系数在0.5654~0.9813之间,表明这些种质材料具有比较丰富的遗传多样性.不同苦丁茶冬青种质材料其不同的起源地域可以从聚类图中清楚地反映出来.RAPD分子标记的结果可以作为判断苦丁茶冬青起源地域的重要参考依据,并可用于探讨苦丁茶冬青不同种质材料的遗传差异和亲缘关系.
【Abstract】 Taking 15 wild germplasm materials of Ilex kudingcha C.J.Tseng as CK,the origins of 26 cultivated materials of I.kudingcha C.J.Tseng were studied via RAPD-PCR analysis.16 random primers with abundant polymorphism were selected from 40 primers tested.These 16 random primers were used for the amplification on the 41 germplasm mentioned above,and 238 bands were obtained.Of which 190 band were polymorphic bands,amounting to 79.8% of the total bands amplified.The Jaccard genetic similarity(GS) of the materials was calculated based on the amplification results and the dendrogram was constructed with UPGMA(unweighted pair group method arithmetic averages).The results showed that the 41 germplasm materials tested were divided into 2 populations,i.e.Population Hainan Island and Population Nanning,Guangxi.The 12 germplasm materials that their precise origins were unknown were clustered into Population Hainan Island,which showed that they originated in Hainan Island.The 12 germplasm materials that their precise origins were unknown were clustered into Population Guanxi-Nanning,which showed that they originated in Nanning region,the Guangxi Zhuang Autonomous Region,or in the neighboring region near Nanning region.The GS of all the 41 materials of I.kudingcha C.J.Tseng tested were distributed between 0.5654-0.9813,which showed that there was abundant considerable genetic diversity.The different origins of the various germplasm materials of I.kudingcha C.J.Tseng tested were clearly showed in the cluster analysis dendrogram.The analysis of RAPD molecular marker could be considered as an important basis for deciding the original region of I.kudingcha C.J.Tseng,and also,it could be used to study the genetic difference and relationship of the various germplasm materials of I.kudingcha C.J.Tseng.
