鼠胚胎干细胞及其衍生神经前体细胞的免疫原性检测
Immunogenicity of Mouse Embryonic Stem Cells and Derived Neural Progenitor Cells
【摘要】 目的:体外观察ES-D3鼠胚胎干细胞(mESCs)及其衍生神经前体细胞(NPCs)的免疫原性。方法:采用无血清培养法从mESCs诱导出NPCs,运用流式细胞仪检测mESCs及NPCs主要组织相容性复合物(MHC)Ⅰ、Ⅱ类分子的组成型表达,及γ干扰素(IFN-γ)诱导作用下MHC分子诱导型表达情况。结果:mESCs在未分化前少量表达MHC-Ⅰ分子但几乎无MHC-Ⅱ分子表达,经诱导分化成NPCs后MHC-Ⅰ分子表达明显上调但MHC-Ⅱ分子仅微弱增加;运用IFN-γ诱导ESCs及NPCs,二者MHC-Ⅰ分子表达进一步增加,而NPCs MHC-Ⅱ分子也呈现相当程度上调。结论:ESCs及其衍生的NPCs具有一定免疫原性,体内移植之后可能引起针对MHC Ⅰ类或Ⅱ类抗原的移植排斥。
【Abstract】 Objective:To study the immunogenicity of embryonic stem cells(ESCs)and derived neural progenitor cells(NPCs) in vitro.Methods: By cultured in serum free selective medium,ES-D3 ESCs were induced into NPCs.The constructive expression of major histocompatibility complex(MHC) class I and II on ESCs and NPCs was evaluated by flow cytometry(FCM) and the inducible expression of MHC-I and II was investigated after IFN-γ induction.Results: ES-D3 ESCs expressed low level MHC-I but no MHC-II molecules.After differentiation the expression of MHC-I on NPCs had a significant up-regulation but MHC-II molecules showed no change.However,when treatment with IFN-γ the ESCs and NPCs demonstrated further moderately increased MHC-I molecules whereas a slight but measurable up-regulation of MHC-II was present on NPCs surfaces.Conclusion: ESCs and their derived NPCs may evoke a possible immune response directed to MHC-I or II molecules on their surfaces.
