丹参注射液对阻塞性黄疸大鼠肠黏膜和细菌移位的影响
Effect of Salvia miltiorrhiga injection on intestinal mucosa and bacterial translocation in rates with acute obstructive jaundice
【摘要】 目的观察丹参注射液对阻塞性黄疸大鼠肠道黏膜和细菌移位的影响。方法SD大鼠30只,雌雄兼用(体质量210~250g),随机分为3组,每组10只:(1)假手术组+生理盐水组(SO+NS组);(2)单纯结扎胆总管+生理盐水组(BDL+NS组);(3)单纯结扎胆总管+丹参注射液组(BDL+SMI组);全部大鼠于实验后第14天处死,处死前1d用吖啶橙标记的大肠杆菌液插管灌胃。大鼠处死后立即:(1)取小肠淋巴结、肝、肾制成匀浆,在荧光镜下观察吖啶橙标记的大肠杆菌并计算细菌移位率;(2)取小肠测定DNA含量;(3)取距回盲部10cm之小肠制备病理切片观察其小肠黏膜厚度和绒毛高度。结果细菌移位率:(1)SO+NS组0%;(2)BDL+NS组63.3%;(3)BDL+SMI组13.3%[(2)vs(3),P<0.05]。小肠DNA含量:(1)SO+NS组(18.33±1.22);(2)BDL+NS组(12.28±1.34);(3)BDL+SMI组(16.87±1.23)[(2)vs(3),P<0.01]。小肠绒毛高度:(1)SO+NS组(489.3±19.4);(2)BDL+NS组(374.6±17.9);(3)BDL+SMI组(465.8±19.2)[(2)vs(3),P<0.01]。小肠黏膜厚度:(1)SO+NS组(678.6±23.8);(2)BDL+NS组(517.5±25.5);(3)BDL+SMI组(597.8±19.8)[(2)vs(3),P<0.01]。结论丹参注射液对阻塞性黄疸大鼠有保护肠黏膜和减少肠道细菌移位的作用。
【Abstract】 Objective To observe the effect of Salvia miltiorrhiga injection(SMI) on the intestine mucosa and baterial translocation(BT) in rats with acute obstructive jaundice(AOJ).Methods Sixty SD rats with male and female mingled and weight from 210 to 250g were randomly divided into three groups,each with 20 rats.SO+NS group: with sham operation,fed with 0.9% normal saline,as the blank control;BDL+NS group: with bile duct ligation alone,fed with normal saline,as the drug control group;BDL+SMI group: with bile duct ligation and SMI intraperitonneal injection,as the experimental group.The animals were killed 10 days after the experiment started.One day before the animals were killed,they were fed with fluid of Eschcrichia coli(E.coli) labeled with acridine by stomach intubation.Mesentary lymph nodes(MLN),liver and kidney were examed with flucororescent microscope for couting the BT rate.DNA contents,thickness of intestinal mucosa and height of intestinal villi were measured.Results(1) Rate of BT were 0% in SO+NS group,63.3% in BDL+NS group and 13.3% in BDL+SMI group,significantly different(P<0.05).(2) DNA contents of intestinal were(18.33±1.22) in SO+NS group,(12.28±1.34) in BDL+NS group and(16.87±1.23) in BDL+SMI group,with significantly difference between the BDL+NS group and BDL+SMI group(P<0.01).(3) Height of intestine villi were(489.3±19.4) in SO+NS group,(374.6±17.9) in BDL+NS group and(465.8±19.2) in BDL+SMI group,with significantly difference between the BDL+NS group and BDL+SMI group(P<0.01).(4) Thickness of intestinal mucosa were(678.6±23.8) in SO+NS group,(517.5±25.5) in BDL+NS group,and(597.8±19.8) in BDL+SMI group,with significantly difference between the BDL+NS group and BDL+SMI group(P<0.01).Conclusion Salvia miltiorrhiga injection could protect the intestinal barrier and prevent the bacteria in intestine from translocation in rats with obstructiue jaundice.
