【摘要】 目的了解白细胞介素-18(IL-18)诱导卵巢癌细胞凋亡的可能性。方法采用荷人卵巢癌HO-8910细胞系的裸鼠皮下移植瘤模型,选择不同剂量(2.5、5、10mg/L)的IL-18进行腹腔内注射(治疗①组IL-18 2.5mg/L,治疗②组IL-18 5mg/L,治疗③组IL-18 10mg/L),对照组为磷酸盐缓冲液(PBS溶液),应用光学显微镜和透射电子显微镜观察形态学变化,Tunel法观察凋亡细胞的变化。结果治疗①组和治疗②组可见凋亡细胞,治疗③组见肿瘤细胞呈片状、条带状坏死区域增多,部分坏死组织呈均质红染无结构区域。IL-18可诱导卵巢癌细胞发生凋亡,不同剂量组的细胞凋亡指数(AI)均明显高于对照组(P<0.01),治疗②组的AI又明显高于治疗①组(P<0.01),治疗①组和治疗③组间差异则无统计学意义(P>0.05)。结论IL-18有诱导卵巢上皮性癌细胞凋亡的作用,这可能是其抗癌机理之一。更多还原

【Abstract】 Objective To investigate the effect of IL-18 on apoptotic induction in ovarian epithelial carcinoma.Methods Twenty-four nude mice with ovarian papillary serous cystadenocarcinoma HO 8910 cell lines were randomly divided into 4 groups,and each group received intraperitoneal injection of IL-18 at various doses(0,2.5,5 and 10 mg/L) for 7 days.The morphology and apoptosis of specimen were observed by light microscopy,transmission electron microscopy(TEM) and TUNEL,respectively.Results There existed apoptotic cells in treatment group I and II.Necrosis occurred in treatment group III.Apoptotic index was significantly higher in treatment group than that in control group(P<0.01),especially in treatment group II,but there was no statistical difference between treatment group I and III(P>0.05).Conclusion IL-18 induced apoptosis in ovarian epithelial carcinoma may be involved in its antineoplasmic mechanism.更多还原
