反相离子对高效液相色谱法测定祖卡木颗粒中吗啡的含量
Determination of morphine content in Zukamu granules by ion-pair RP-HPLC
【摘要】 目的建立祖卡木颗粒中吗啡的含量测定方法。方法以Hypersil BDS C18柱(4.6 mm×250 mm,5μm)为色谱柱,流动相:0.01 mol/L磷酸二氢钾、0.01 mol/L庚烷磺酸钠混合溶液-乙腈-三乙胺(体积比85∶15∶0.03),体积分数为5%的冰醋酸调pH值为4.9;流速:1.0 mL/min;检测波长:240 nm;柱温:30℃;进样量:20μL。结果吗啡的线性范围为5.22~52.2μg/mL,r=0.9999,平均回收率为96.74%,RSD=0.95%。结论本文建立的方法简便,测定结果准确,重复性好,可用于该制剂中吗啡的含量测定。
【Abstract】 Objective To set up an ion-pair RP-HPLC method for determining the content of morphine in Zukamu granules.Methods The analysis was performed on an Hypersil BDS C18 column(4.6 mm×250 mm,5 μm).The mobile phase was a mixture of 0.01 mol/L potassium dihydrogen phosphate and 0.01 mol/L sodium heptanesulfonate-acetonitrile-triethylamine(85∶15∶0.03),adjusted with 5% glacial acetic acid to pH 4.9.The flow rate was 1.0 mL/min and the detection wavelength 240 nm.Results The linear range of morphine was 5.22~52.2 μg/mL(r=0.9999).The average recovery was 96.74% with RSD 0.95%.Conclusion This method is simple,accurate and reproducible and can be used for the determination of morphine content in Zukamu granules.
