【摘要】 目的探讨Survivin和Caspase-3在胆管细胞癌中的表达及相互关系。方法用免疫组化S-P法分别检测Survivin和Caspase-3在胆管细胞癌、癌旁组织及正常胆管组织中的表达。结果①Survivin在胆管细胞癌中的表达显著高于癌旁组织和正常胆管组织(P<0.01);Caspase-3在胆管细胞癌中的表达显著低于癌旁组织和正常胆管组织(P<0.01)。②在胆管细胞癌中,Survivin的表达随着肿瘤分化程度的降低而增强(rs=0.404,P<0.01),Caspase-3的表达随着肿瘤分化程度的降低而减弱(rs=-0.389,P<0.05)。③Survivin和Caspase-3在胆管细胞癌中的表达呈负相关(rs=-0.765,P<0.01)。结论Survivin的高表达和Caspase-3的失表达抑制了胆管细胞癌细胞的凋亡,可能与胆管细胞癌细胞的生长有关;Survivin和Caspase-3表达的负相关性反映了Survivin抑制Caspase-3介导的细胞凋亡,可能与癌细胞的发生发展有密切关系。更多还原

【Abstract】 Objective To investigate expression of Survivin and Caspase-3 in cholangiocarcinoma and their relationship.Methods Immunohistochemical assay(streptavidin-biotin peroxidase method) was used to detect the expression of Survivin and Caspase-3 in 42 cholangiocarcinoma tissues,20 adjacent noncancerous bile duct lesions and 11 benign bile duct lesions.The chi-square test and association analysis were used to evaluate the difference of significance.The SPSS12.0 software package was used to conduct statistical analysis.The P value <0.05 was thought as statistical significant.Results ①Expression of Survivin was significantly higher in cholangiocarcinoma(61.9%) than that in adjacent noncancerous(5%) and benign bile duct lesions(0%)(P<0.01).Expression of Caspase-3 was significantly lower in cholangiocarcinoma(45.2%) than that in adjacent noncancerous(85.0%) and benign bile duct lesions(90.9%)(P<0.01).②Expression of Survivin in cholangiocarcinoma tissues increased with the reduction of the differentiation level of tumor(rs=0.404,P< 0.01),whereas expression of Caspase-3 in cholangiocarcinoma tissues decreased with the reduction of the dif-ferentiation level of tumor(rs=-0.389,P<0.05).③The negative correlativity between the expression of Survivin and Caspase-3 could be found with statistical significance(rs=-0.765,P<0.01).Conclusion High expression of Survivin and low expression of Caspase-3 depress the apoptosis of cholangiocarcinoma,which maybe related to the growth of cholangiocarcinoma.The negative correlativity of the expression between Survivin and Caspase-3 suggests that Survivin inhibit the apoptosis induced by Caspase-3,which maybe related to the occurrence and development of cholangiocarcinoma.更多还原
