马铃薯试管薯诱导体系的优化
Optimization of the micro-tuber inducement of potato
【摘要】 以马铃薯脱毒苗为材料,探讨了不同蔗糖浓度、光照条件、固液培养方式和不同浓度梯度外源细胞分裂素6-BA、ZT和KT对试管薯诱导率的影响。结果表明:以MS+10%蔗糖(pH 5.8)为基本培养基,在黑暗条件下加入一定量的外源细胞分裂素,有利于试管薯的形成和产量的提高,其中以培养基MS+10%蔗糖+5 mg/LKT的诱导率最高(100%)且试管薯质量最好。
【Abstract】 Virus-free plantlets of Solanum tuberosum L.was used as materials,MS+10% sucrose(pH 5.8)was used as basic substrate to study the influence of different concentration gradient exogenous Cytokinins 6-BA、ZT and KT on micro-tuber induction of potato under the condition of darkness.The results show that adding a certain amount of exogenous Cytokinins was in favor of the formation of micro-tuber and the advance of the yield;The best medium for inducement and growth of micro-tuber of Solanum tuberosum L.is MS+10% sucrose+ KT 5 mg/L,with which the micro-tuber induction rate could reach to 100% and micro-tuber grew vigorously.
