阿尔茨海默病患者外周血TNF水平及TNFβG252A基因多态性

【摘要】 目的探讨肿瘤坏死因子TNFβG252A基因多态性与阿尔茨海默病的关系。方法用放免法测定TNF水平,运用多聚酶链反应技术检测66例阿尔茨海默病及143例正常人TNFβG252A基因多态性。结果AD组肿瘤坏死因子TNFβG252A1/1、1/2、2/2表型频率分别为0.242、0.591、0.167,对照组分别为0.252、0.573、0.175;AD组TNFβG252A1、TNFβG252A2基因频率分别为0.5379、0.4621,对照组分别为05385、0.4615,TNFβG252A各种基因型频率在患者组与正常对照组之间的差异无统计学意义(P>0.05),而血浆中TNF水平,AD组明显高于对照组,两组之间比较差异有统计学意义(P<0.05)。结论血清TNF水平显著升高,提示炎性反应在阿茨海默病病程中有重要作用,TNFβG252A基因多态性与AD无明显相关。

【Abstract】 Objective To investigate the relationship between plasma levels of tumor necrosis factor(TNF) or its polymorphism and Alzheimer’s disease. Methods Plasma level of TNF was measured by RIA,and the polymorphism of TNFβG252A gene was screened by PCR-RELP. Results The gene frequency of TNFβG252A in AD group was as followed; TNFβG252A 1/1, 1/2, 2/2 genotype frequency: 0.242, 0.591, 0.167; TNFβG252A 1 and TNFβG252A 2 genefrequency: 0.252, 0.573, 0.175. The gene frequency of TNFβG252A in control group was as followed; TNFβG252A 1/1, 1/2, 2/2, genotype frequency: 0.252, 0.573, 0.175,TNFβG252A 1, TNFβG252A 2 genefrequency: 0.9021, 0.0979. There was no significant differences in the frequencies of TNFβG252A gene between the patients and control group(P>0.05). Alzheimer’s group showed significantly higher plasma level of TNF than control group(P<0.05). Plasma TNF increased in AD group. Conclusion TNF play important role in the process of AD and no evident association is found between TNFβG252A polymorphism and Alzheimer’s disease.
