转sbk和sck抗虫基因水稻的获得
Obtaining Transgenic Rice Plants Harboring sbk and sck Insecticidal Genes
【摘要】 转入多个抗虫基因是解决水稻抗虫的一条有效途径。本研究采用根癌农杆菌介导法将2个抗虫基因(sbk和sck)同时转入3个水稻品种中,通过一系列的分子检测(PCR和RT-PCR)分析,这2个基因已经成功转入3个水稻品种中并得到了表达;同时还获得1株不含抗生素筛选标记基因(hpt)和抗虫sbk基因,却含抗虫sck基因的转基因植株,命名为YJ27-5sck。用该植株作为供体,通过杂交将sck抗虫基因转育到另外5个水稻品种中且同样得到了表达,获得了不含筛选标记基因的转sck基因阳性植株。
【Abstract】 It might be one of the effective ways to solve the problems of rice insect harm by introducing multigenes into rice. In this research, two insecticidal genes (sbk and sck) have been transferred into three rice varieties mediated by Agrobacterium tumerfaciens approach, the results of PCR identifications indicated that the two genes have been integrated into the genomes of tested three rice varieties and the targeted genes would be successfully expressed. And also we distinguished a transgenic individual which its antibiotics select marker gene (hpt) and sbk insecticidal gene were negative but the sck insecticidal gene was positive, named as YJ27-5sck. By using the YJ27-5sck as insecticidal gene donor, we made some crosses with five other varieties, RT-PCR assay indicated that the sck gene might be introduced into their genomes and well expressed. This research might provide the transgenic germplasms for further transgenic breeding program.
