【摘要】 目的:了解内蒙古包头地区汉族人群IgA肾病患者肿瘤坏死因子α(TNF-α)-308位点基因型的分布特点。方法:应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测97例IgA肾病患者和73例正常人的TNF-α基因型。结果:IgA肾病患者组中TNF-α基因G/A多态GG、GA、AA基因型频率分别为85.6%、11.3%和3.1%,其中G和A等位基因频率分别为91.2%和8.8%。正常对照组中,TNF-α基因G/A多态GG、GA、AA基因型频率分别为91.8%、5.5%和2.7%,其中G和A等位基因频率分别为94.5%和5.5%。IgA肾病患者组与正常对照组比较,基因型频率差异有显著性(χ2=27.2,P<0.05),等位基因频率之间差异无显著性(χ2=0.659,P>0.05)。结论:TNF-α-308位点基因多态性与内蒙古包头地区汉族人群IgA肾病的发病有相关性,等位基因频率之间差异无显著性(χ2=0.659,P>0.05)。更多还原

【Abstract】 Objective To study the relationship between tumor necrosis factor-α(TNF-α) gene-308 site polymorphism and IgA nephropathy in ethnic Han population in Bao tou region of Inner Mongolia.Methods The polymorphism of TNF-α gene was studied with PCR-RFLP in 97 patients with IgA nephropathy and 73 controls.Results The frequencies of TNF-α gene genotypes CG,GA,AA were 85.6%,11.3% and 3.1% respectively in patients with IgA nephropathy.The frequencies of alleles G and A were 91.2% and 8.8%.The frequencies of TNF-α gene genotypes CG,GA,AA were 91.8%,5.5% and 2.7% in controls. The frequencies of alleles G and A were 94.5% and 5.5%.The distribution of G/A polymorphism of TNF-α gene was in agreement with Hyrdy-Weinberg equilibrium.The frequencies of TNF-α gene genotypes in patients with IgA nephropathy were significantly different(χ2=27.2,P<0.05) from those in the controls.The frequencies of alleles G and A were not significantly different(χ2=0.659,P>0.05) from those in the controls.Conclusion TNF-α gene-308 polymorphism is associated with IgA nephropathy,A allele may not be a risk factor in the development of IgA nephropathy in ethnic than population in Bao tou region of Inner Mongolia.更多还原
