【摘要】 目的分析环氧合酶-2(COX-2)在肝细胞癌(HCC)中的表达及意义。方法应用改良的手工制备组织芯片方法制备含150例肝细胞癌、92例癌旁肝硬化、45例癌旁慢性肝炎以及37例肝内转移灶和13例正常肝组织的组织芯片,结合免疫组织化学染色和图像分析技术检测COX-2蛋白的相对表达量。结果COX-2蛋白在HCC表达显著低于癌旁肝硬化、癌旁慢性肝炎(P<0.01),但高于正常肝组织(P<0.05),差别具有统计学意义。在HCC组织中,低分化HCC的COX-2表达量显著低于高分化的HCC,差别有统计学意义(P<0.01)。COX-2蛋白的表达与性别、肿瘤大小、组织学类型、包膜侵犯及转移均无关(P>0.05)。结论COX-2在HCC的发生中发挥重要作用,并与HCC的分化程度密切相关,但与HCC进展及侵袭转移关系不大。更多还原

【Abstract】 Objective To detect the expression of cyclooxygenase-2(COX-2) in hepatocellular carcinoma(HCC). Methods Tissue chips were constructed by modified manual tools,which contained 150 HCC samples,92 cirrhotic livers adjacent to HCC samples,45 chronic hepatitis livers adjacent to HCC samples,13 normal liver tissue samples and 37 intrahepatic metastasis lesion samples. The expression of COX-2 was determined by immunohistochemistry and semi-quantitative image analysis. Results The expression of COX-2 in HCC was lower than that in cirrhotic liver adjacent to HCC tissue or chronic hepatitis tissue,but higher than that in normal liver tissues(P<0.01). More lower differentiation of HCC more the expression of COX-2 decreased. COX-2 expression was not correlated with sex,age,tumor size,histological type,tumor invasion and metastasis(P>0.05). Conclusion COX-2 might play an important role in the process of HCC tumor genesis and the differentiation of HCC,but not related with invasion and metastasis.更多还原
