抗口蹄疫病毒siRNA在HEK-293细胞诱导β-干扰素的研究
Research on IFN-β Activation in HEK-293 cell by siRNA Targeting Foot-and-Mouth Disease Virus
【摘要】 利用shRNA(short-hairpin RNA)表达质粒在HEK-293细胞上评估了多个dsRNA诱发产生干扰素(IFN)的能力.结果显示,p3D3(19 nt),pPol(56 nt),p21NT(21 nt)和p63NT(63 nt)能够诱发IFN-β的产生,其中p3D3和p63NT能强烈诱导IFN-β;而p3D1(19 nt),p3D2(19 nt)和pLacZ(83 nt)没有明显的IFN-β诱发能力.结果显示,诱发干扰素产生的能力与dsRNA的片段长度没有关系,可能与其序列有一定相关性.
【Abstract】 Several plasmids which can express short-hairpin RNAs(shRNAs) were constructed to evaluate their potentials of activating IFN system in HKE-293 cells.The results showed that shRNA expression plasmids p3D3(19 nt),pPol(56 nt),p21NT(21 nt) and p63NT(63 nt) could induce interferon-β expression,especially p3D3 and p63NT exhibited great potentials of interferonβ activation.While p3D1(19 nt),p3D2(19 nt) and pLacZ(83 nt) did not show marked potentials of inducing interferon-β expression.The results suggested that the ability to activate interferon expression had no relations with the length of dsRNA,but was more likely to be sequence-dependent.
