糖尿病大鼠肠道、胰腺GLP-1受体mRNA的表达
Expression of GLP-1 receptor mRNA in intestine and pancreas of diabetic rats
【摘要】 目的:研究胰高血糖素样多肽1(GLP-1)受体mRNA表达水平在大鼠肠道不同部位和胰腺分布的特点及两者间的差异.方法:以Wistar大鼠为对照,口服葡萄糖耐量试验确定自发性2型糖尿病模型GK大鼠已发病,采用实时定量PCR方法(SYBR Green法)检测大鼠不同肠段和胰腺的GLP-1受体mRNA表达水平.结果:GLP-1受体mRNA表达中,正常大鼠不同肠段以空肠表达量最低,与回肠和结肠相比较差异有统计学意义(P<0.05),回肠与结肠相比较差异无统计学意义(P>0.05).糖尿病组大鼠空肠GLP-1受体mRNA相对表达量为0.27,而对照组大鼠空肠的相对表达量为0.59;糖尿病组大鼠结肠GLP-1受体mRNA相对表达量为0.5,对照组大鼠结肠的相对表达量为1;糖尿病组大鼠胰腺GLP-1受体mRNA相对表达量为0.37,而对照组大鼠胰腺的相对表达量为0.97,两组间空肠、结肠和胰腺GLP-1受体mRNA相对表达量差异均有统计学意义(P<0.05).糖尿病组回肠GLP-1受体mRNA相对表达量为0.73,对照组回肠的相对表达量为0.86,两组间差异无统计学意义(P>0.05).结论:糖尿病大鼠GLP-1受体mRNA相对表达量在空肠、结肠和胰腺的下调,提示空肠、结肠和胰腺GLP-1受体的表达变化可能与GK大鼠的糖尿病发生发展有关.
【Abstract】 AIM: To observe the distribution and difference of the expression of glucagon like peptide-1(GLP-1) receptor mRNA in different parts of intestine and pancreas in rats.METHODS: Four Wistar rats were enrolled into control group.Oral glucose tolerance test was used to identify the spontaneous type 2 diabetic GK rat models.Real-time quantitative PCR(SYBR Green) was done to detect the expression of GLP-1 receptor mRNA in different parts of intestine and pancreas in rats.RESULTS: The jejunum had the lowest expression level of GLP-1 receptor mRNA compared with ileum and colon in normal rats(P<0.05),and there was no difference in GLP-1 receptor mRNA expression level between ileum and colon(P>0.05).Relative expression quantity of GLP-1 receptor mRNA in jejunum of diabetic rats was 0.27,while it was 0.59 in control group;GLP-1 receptor mRNA relative expression quantity in colon of diabetic group was 0.5,while it was 1 in control group;the relative expression quantity in pancreas of diabetic group was 0.37,while 0.97 in control group;the differences of GLP-1 receptor mRNA expression were statistically significant between the 2 groups in jejunum,colon and pancreas(P<0.05).GLP-1 receptor mRNA relative expression quantity in ileum of diabetic group was 0.73,while the quantity was 0.86 in control group,with no statistical difference between the 2 groups(P>0.05).CONCLUSION: The downregulation of GLP-1 receptor mRNA expression in the jejunum,colon and pancreas in diabetic rats suggests that the changes in the expression of GLP-1 receptor mRNA in jejunum,colon and pancreas may be related to the development of diabetes in GK rat.
