HSP90在离体人真皮成纤维细胞热损伤中的保护作用
Protective effect of HSP90 against heat injury on human dermal fibroblasts in vitro
【摘要】 目的:探讨人真皮成纤维细胞热损伤变性后HSP90的保护作用.方法:培养的人真皮成纤维细胞在52℃,30s热水损伤24h后用免疫荧光观察HSP90表达部位的改变并与正常组比较,用Western Blot进一步分析正常的FB和热变性的FB在3,6,12,24,48,72,96及120h时相点HSP90表达量的改变.结果:用免疫荧光观察到FB热损伤后24h胞质胞核中出现HSP90高表达.Western Blot显示3h即有HSP90表达量升高,24h达到高峰,维持到96h,120h恢复到正常.结果:FB热变性可以引起HSP90异常高表达,HSP90对FB热损伤具有保护作用.
【Abstract】 AIM:To investigate the protective effect of HSP90 against heat injury on human dermal fibroblasts in vitro.METHODS:Fibroblasts were impaired by heating under water at 52℃ for 30 s.The expression of HSP90 in both the normal group and the experimental group was observed by immunohistochemistry after 24 h.Meanwhile,we also detected the expression level of HSP90 in the experimental group by Western Blot on 3,6,12,24,48,72,96 and 120 h.RESULTS:The expression of HSP90 in the experimental group was obviously stronger than that in the normal group after 24 h.The expression level of HSP90 in the experimental fibroblast began to increase on 3 h,peaked on 24 h,retained for 96h and restored to the normal on 120 h.CONCLUSION:Due to heat injury,abnormal high expression of HSP90 in the experimental fibroblast is found.HSP90 may protect fibroblasts from heat injury.
