大鼠海马中56KD、83KD蛋白诱导神经干细胞向神经元分化的协同作用研究

【摘要】 目的:探讨切割海马伞大鼠海马中56KD、83KD蛋白在诱导神经干细胞向神经元分化方面是否具有协同作用。方法:切割海马伞后的海马进行非变性聚丙烯酰胺凝胶电泳,取低分子量区的56KD、65KD和83KD 3条差异蛋白条带电洗脱后以不同的组合加入鼠胚神经干细胞的分化培养液中诱导神经干细胞分化,12天后行MAP-2免疫荧光染色,记数其中MAP-2阳性神经元数,胞体面积和细胞周长。用STATA7.0软件进行方差分析和两两比较。结果:56KD+65KD+83KD组和56KD+83KD组MAP-2神经元的数量是47.1±9.64,43.7±8.41,胞体大,突起丰富;65KD+83KD组、56KD+65KD组、56KD组和83KD组MAP-2神经元的胞体较小,突起较短;65KD组和对照组的神经元胞体最小,突起最短。MAP-2神经元的数量、面积和周长统计分析结果显示:56KD+65KD+83KD组和56KD+83KD组与其他6组均有显著性差异。56KD+65KD+83KD组和56KD+83KD组之间没有显著性差异;65KD+83KD组、56KD+65KD组、56KD组和83KD组四组之间没有显著性差异,但是它们与65KD组和对照组均有显著性差异,65KD组和对照组之间没有显著性差异。结论:56KD、83KD蛋白在诱导神经干细胞向神经元分化方面具有协同作用。

【Abstract】 Objective:To explore the Synergism of the 56KD and 83KD Protein in Hippocampi on Inducing NSCs differentiating into neurons.Method:Native-PAGE was carried out in the hippocampi with the fimbria transection,Then the differential protein of 56KD,65KD and 83KD were electroeluted,divided into 8 groups and co-cultured the NSCs clone spheres of rats.On the 12th day after culture,MAP-2 immunofluorescence was used to detecte the differentiation of NSCs into neurons.The numbers,dimensions and perimeters of the MAP-2 positive neurons were analyzed with image analysis system.The variance and comparisons of every two groups were employed with the software of STATA 7.0.Results:the numbers of MAP-2 positive neurons in 56KD+65KD+83KD group and 56KD+83KD group were 47.1±9.64 and 43.7±8.41,the sizes of cells were big,the processes were abundant,MAP-2 positive neurons in 65KD+83KD group,56KD+65KD group,56KD group and 83KD group,the sizes of ceils were small and the processes were short;That in 65KD group and control group the smallest and the processes were the shortest.Statistic results showed that for the numbers, dimensions and perimeters of MAP-2 positive neurons,there were significant differences among 56KD +65KD +83KD,56KD +83KD groups and other 6 groups,but there weren’t significant differences between 56KD+65KD+83KD group and 56KD+ 83KD group,and there weren’t differences among 65KD+83KD group,83KD group,56KD+65KD group and 56KD group,but there were differences among 65KD group,control group and other groups,there weren’t significant differences between 65KD group and control group. Conclusions:There were synergism of the 56KD and 83KD protein in hippocampi on inducing NSCs differentiating into neurons;
