白桦长纤维性状ISSR和SCAR标记的分析

【摘要】 通过对天然白桦群体583株个体的纤维长度进行测定,选取其中有代表性的100株个体,利用简单重复序列间区标记技术(Inter-simple sequence repeat,ISSR)进行基因组差异分析,通过扩增条带与性状表现间的多元回归分析,筛选出与白桦纤维长度显著相关的分子标记。经过20个ISSR引物筛选,有4个引物的4条片段与纤维长度呈极显著相关,其中片段"BFLI-3"与纤维长度的回归系数为-0.331,显著性达到1%水平。对此片段进行克隆、测序后,成功转化成与长纤维性状相关的序列特征化扩增区域(Sequence characterized amplified region,SCAR)标记,此标记对短纤维白桦的鉴定效率达76%。

【Abstract】 After measuring fiber length trait (FLT) of 583 individuals from nature birch population, 100 individuals were selected as representative variety for correlated fragment screening with Inter-simple sequence repeat (ISSR) technique. Four fragments from four primers were identified having significant correlation with FLT based on multiple regression analysis. The correlation coefficient between BFLI-3 and FLT was -0.331, showing a significant difference at the 0.01 level. BFLI-3 was cloned, sequenced and transformed into Sequence Characterized Amplified Region (SCAR) marker. The identifying percentage of short-fiber birch by this SCAR marker reached 76%.
