长白落叶松愈伤组织诱导与不定芽分化

【摘要】 以长白落叶松成熟胚作为外植体进行消毒和不定芽诱导。研究结果表明:将去皮种子用流水冲洗3d,用70%酒精消毒1min,转入1%的次氯酸钠消毒50min后,再将剥出的胚放入0.5%的次氯酸钠中浸泡2s为最佳消毒方法;在光照条件下(约1600lx),BM培养基中填加1.0mg/LBA,愈伤组织与不定芽的诱导效果最好,愈伤组织诱导率达到100%,不定芽分化率为73.8%;在BM培养基中加入1%活性碳对不定芽有伸长作用。

【Abstract】 Mature embryos of Larix olgensis as explants were disinfected and then used to induce adventitious bud. The optimum disinfection method was that after rinsed in running water for three days, the seeds without capsule were surface decontaminated with 70% ethanol for 1minute followed by disinfected with 1% sodium hypochlorite for 50 minutes, and then mature zygotic embryos were removed from the seeds and immersed in 0.5% sodium hypochlorite for 2 seconds. The best induction conditions were obtained as BM medium supplemented with 1.0mg/L BA under a light intensity of 1600lx. The callus induction and the differentiation frequency of adventitious buds reached 100% and 73.8%. 1%, respectively. Activated carbon added to BM medium could enhance adventitious bud elongation.
