60Coγ诱变对白桦愈伤组织再生的影响及再生植株染色体数量变异分析

【摘要】 以白桦(Betula platyphylla)叶片愈伤组织为材料,对诱变后的愈伤组织分化、不定芽生根情况以及染色体数量变异进行了研究:经γ射线处理后,在10~30 Gy范围内愈伤组织的不定芽分化率随着辐射剂量的增加显著降低;染色体数量变异在10~25 Gy呈上升趋势。试验确定,愈伤组织的适宜剂量为20 Gy,该剂量的愈伤组织分化率为53.2%,染色体数量变异率为80.3%。10~30Gy剂量的γ射线处理后,愈伤组织再生植株染色体数量变异率均在79.5%以上,多数产生亚二倍体变异类型。当辐射剂量超过25Gy时,再生植株基本都变成了亚二倍体,γ射线对照射后的白桦不定芽生根具有推迟和抑制作用。

【Abstract】 The differentiation of induced leave callus by gamma ray,rooting of adventitious bud and variation in chromosome number were studied using Betula platyphylla leaf callus as testing materials.Differential rate of callus reduces greatly with radiation dose increasing within 10 Gy and 30 Gy and exhibits an increasing tendency within 20 Gy and 25 Gy.Result shows that the optimal radiation dose is 20 Gy,and the differential rate of callus under this dose is 53.2% and the mutation probability of chromosome number is 80.3%.Within a dose range of 10 Gy to 30 Gy,the mutation probability of chromosome number of the regenerated plants is above 79.5%,and most of them appear to be hypodiploid.When the dose is more than 25 Gy,the regenerated plant mostly may turn to be hypodiploid.Gamma ray could postpone and restrain the rooting of adventitious bud of B.platyphylla.
