H5亚型AIVHA和HA1的Sf-9昆虫细胞表达及其作为检测抗原的初步比较分析

【摘要】 利用杆状病毒表达系统,在昆虫细胞Sf-9中分别表达了H5亚型禽流感病毒的HA、HA1。相同表达条件下,Western Blot分析比较表明,HA、HA1均与H5亚型AIV抗血清反应,且不与H7、H9亚型AIV抗血清发生交叉反应,具有良好的特异性;以Western Blot检测鸡、鸭AIV抗血清,初步表明,重组杆状病毒表达的HA、HA1均可作为H5亚型AIV血清检测的抗原,HA优于HA1。

【Abstract】 HA and HA1 of H5 subtype AIV were expressed via baculovirus system in Sf-9 insect cells. The recombinant proteins were analyzed and compared by Western Blot under the same expression condition. They were recognized by antiserum against H5 but not antiserum against H7 or H9 subtype AIV. Briefly study on the detection of ducks and chicken anitsera to H5 AIV using these two recombinant proteins showed that HA and HA1 had the potential as detective antigen for the detection of duck and chicken antisera against AIV, and HA was better than HA1.
