5-杂氮脱氧胞苷对JEG-3细胞侵袭力的影响及相关机制探讨
Effect of 5-aza-2′-deoxycytidine on the invasion of human JEG-3 choriocarcinomacells and preliminary study on related mechanism
【摘要】 目的研究甲基化酶抑制剂5-杂氮脱氧胞苷(5-aza-2-′deoxycytidine,ADC)对人绒毛膜上皮癌细胞株JEG-3细胞侵袭能力及其对JEG-3细胞H19甲基化状态的影响,从表观遗传学角度探讨滋养细胞侵袭行为的相关调控机制。方法Matrigel侵袭小室模型观察ADC处理JEG-3细胞24h后细胞侵袭能力的变化。亚硫酸氢盐修饰后测序法检测ADC对JEG-3细胞中H19基因甲基化的影响。结果经100μmol/L ADC作用24h后的JEG-3细胞侵袭能力明显降低,较对照组差异有统计学意义(P<0.01)。经ADC作用后的JEG-3细胞中H19基因甲基化程度降低。结论甲基化酶抑制剂ADC可降低H19基因甲基化程度,并抑制滋养细胞侵袭能力。H19甲基化状态改变可能参与调控滋养细胞侵袭能力。
【Abstract】 Objective To explore the methylation regulation of 5-aza-2′-deoxycytidine(ADC) on H19 gene and its effect on JEG-3 cell line invasion.Methods Transwell cell culture inserts were used to detect the invasion of JEG-3 cells after treated with ADC.H19 methylation level was detected by bisulfite PCR and sequencing.Results After ADC treatment,the invasion ability of JEG-3 cells was depressed.H19 mathylation level was decreased.Conclusion ADC could suppress invasive ability of choriocarcinoma cells and inhibit H19 methylation.
