针刀疗法对腰三横突征模型大鼠局部组织IL-6、IL-10、TNF-α、TGF-β的影响

【摘要】 目的:观察第三腰椎横突综合征模型大鼠局部组织相关细胞因子的含量变化,探讨针刀松解法治疗机制。方法:选用SD大鼠28只,随机分为正常组、模型组、电针组、针刀组,后三组首先用在一侧第三腰椎横突尖处放置明胶海绵的方法建立第三腰椎横突综合征的动物模型,14天后,后两组分别给予6次电针或2次针刀松解干预治疗。造模56天后,取大鼠第三腰椎横突周围局部组织,采用放射免疫法检测局部组织IL-6、IL-10、TNF-α、TGF-β含量。结果:模型组大鼠IL-6、IL-10、TNF-α、TGF-β含量都明显升高(P<0.01);针刀组IL-6、IL-10、TNF-α、TGF-β含量较模型组降低,电针组IL-6、IL-10含量降低(P<0.01,P<0.05)。结论:针刀松解法可以减轻局部组织内炎性细胞因子的产生,这可能是针刀治疗软组织损伤、改善局部组织瘢痕粘连状况疗效更优的原因之一。

【Abstract】 Objective: To observe the change of cytokine in local tissue of rats with the third lumbar transverse process syndrome, and to study the therapeutic mechanism of needle-knife dissolution. Methods: Twenty-eight SD rats were randomly divided into normal control group, model group, needle-knife dissolution group and electroacupuncture (EA) group. The model rats with the third lumber transverse process syndrome were established by inserting gelfoam into the deep subfascial part of the 3rd lumbar transverse process. 14 days later, needle-knife dissolution group and EA group were treated with electroacupuncture 6 times and needle-knife dissolution 2 times respectively. Scars and adhesion and L-6, IL-10, TNF-α, TGF-β contents in local tissue were detected with radioimmunoassay on the 57th day. Results: In model group, L-6, IL-10, TNF-α, TGF-β contents increased significantly (P<0.01), inflammatory reaction was intense, the scars and adhesion were formed obviously. In needle-knife dissolution group, L-6, IL-10, TNF-α, TGF-β contents decreased significantly (P<0.01, P<0.05). In EA group, L-6, IL-10 contents decreased (P<0.01, P<0.05). Inflammatory reaction, the scars and adhesion of the two groups were not obvious. Conclusion: Needle-knife dissolution can decrease the production of the inflammatory cytokine, reduce the damage of inflammatory reaction to local tissue. This may be the reason why needle-knife dissolution was more effective on reducing the scars and adhesion in local tissue.
