【摘要】 目的:探讨结直肠癌c-erbB-2癌基因扩增的临床意义。方法:应用差别PCR技术检测52例结直肠癌患者在c-erbB-2癌基因的扩增情况。结果:48.1%(25/52)的结直肠癌发现有c-erbB-2癌基因的扩增;有淋巴结转移及Dukes分期C+D期患者c-erbB-2基因扩增率(72.0%及68.0%)分别高于无淋巴结转移及Dukes分期A+B期患者(25.9%及29.6%);c-erbB-2基因扩增与肿瘤组织学分化和组织病理类型显著相关;伴有腺瘤性息肉的结直肠癌患者c-erbB-2基因扩增率高于不伴有腺瘤性息肉的结直肠癌患者(100.0%对43.5%,P<0.05)。结论:差别PCR技术检测c-erbB-2癌基因的扩增是一种快速简便、灵敏可靠的方法;c-erbB-2癌基因的扩增在一定程度上反映结直肠癌的分化状态和生物学行为,对结直肠癌的预后和转移潜力是一种良好的判断指标,并可指导临床治疗。更多还原

【Abstract】 Objective:To explore the clinical significance of c-erbB-2 oncogene ampilification in colorectal cancer(CRC).Methods: C-erbB-2 oncogene ampilification was examined by differential polymerase chain reaction(dPCR) in 52 cases of colorectal cancer.Results: C-erbB-2 oncogene ampilification was found in 48.1%(25/52)of colorectal cancer.The ampilification incidence of c-erbB-2 in patients with lymph node metastasis and in Dukes’stage C+D(72% and 68%) was higher than that in patients without lymph node metastasis and in Dukes’stage A+B(25.9% and 29.5%),respectively(both,P<0.01).Amplification of c-erbB-2 gene was correclated significantly with histological grade(P<0.01) and histopathological phentypes(P<0.05).Occurrence rate of c-erbB-2 oncogene amplification in the patients of colorectal cancer with adenomatous polyp was higher than that in those without adenomatous polyp(100% vs 43.5%,P<0.05).Conclusion: dPCR is a quick,simple,sensitive and reliable method to detect c-erbB-2 0ncogene amplification.To some extent,the amplification of c-erbB-2 oncogene represents the differentiation and biological behavior of colorectal cancer,thus can be taken as an independent prognostic marker for predicting the prognosis and metastatic potantial of colorectal cancer can be used to guide the clinical treatment of the disease.更多还原
