TTX停搏液对离体鼠心结蛋白作用及机制的研究
Effects and mechanisms of cardioplegia with tetrodotoxin on desmin of isolated rat hearts
【摘要】 目的研究Na+通道阻滞剂河豚毒素(tetrodotoxin,TTX)停搏液对离体鼠心结蛋白的作用及机制。方法20只Wistar大鼠随机分为实验组和对照组,建立离体鼠心Langendorff和Neely灌注模型,搏动稳定后,灌注30min后停搏60min,再复灌60min,观察各组心脏停搏前后心功能指标的变化,通过免疫组化检测钙蛋白酶、免疫组化和蛋白印迹(Western blotting,WB)检测结蛋白的分布及量的变化。结果复灌后,实验组的各项心功能恢复率明显优于对照组(P﹤0.01或P﹤0.05),钙蛋白酶分布整齐,表达量为0.07156±0.00005,明显低于对照组的0.14592±0.00003(P﹤0.01);结蛋白分布紊乱,表达量为574.1±4.7,明显高于对照组的383.5±8.6(P﹤0.01)。结论TTX停搏液对离体鼠心肌缺血-再灌注损伤(myocardial ischemia/reperfusion injury,MIRI)时的结蛋白及心功能有保护作用,其机制可能是通过抑制钙蛋白酶的激活而引起的。
【Abstract】 Objective To investigate the effects and mechanisms of cardioplegia with Na+ channel inhibitor tetrodotoxin(TTX) on myocardial desmin of isolated rat hearts. Methods While Langendorff and Neely perfusion models were steadily established for 30 minute 20 rat hearts(10 hearts in each group randomly) were respectively arrested by TTX cardioplegia solution in Group TTX, and arrested by St. Thomas No.2 solution(STH-2) in Group STH-2. These arrested hearts remained for 60 minutes and then underwent reperfusion for 60 minutes. Pre-ischemia and post-ischemia hemodynamic parameters of the rat hearts were studied and distribution of myocardial desmin was detected by Immunohistochemistry and volume change of myocardial desmin was detected by Westem Blottimy(WB), calpain was detected by Immunohistochemistry. Results There were no difference in myocardial hemodynamic parameters between two groups before myocardial ischemia, but after reperfusoin the outcome in Group TTX was better than that in Group STH-2(P﹤0.01 or P﹤0.05). Expression of myocardial desmin in Group STH-2 decreased conspicuously when calpain grew(P﹤0.01 ). Conclusions Cardioplegia with TTX has protective effects on myocardial desmin and heart function in MIRI rats and it works by lowering activity of caplain.
