巨细胞病毒感染对神经细胞[Ca2+]i/钙调素及线粒体膜电位的影响
Effect of [Ca2+]i and mitochondria transmembrane potentials in cmv infected neuronal cells
【摘要】 目的观察巨细胞病毒感染组和正常对照组小鼠海马神经细胞[Ca2+]i CaMmRNA及线粒体膜电位。方法将BALB/C乳鼠同母鼠随机分为实验对照组(18只)和病毒感染组(35只),感染后56天检测两组小鼠海马神经细胞的[Ca2+]i CaMmRNA及线粒体膜电位。结果病毒感染组小鼠海马[Ca2+]i CaMmRNA增高及线粒体膜电位明显下降(P<0.01),与正常对照组比较差异有显著性。结论巨细胞病毒感染所引起的小鼠海马[Ca2+]i CaMmRNA及线粒体膜电位的改变可能是其导致神经系统功能紊乱的机制之一。
【Abstract】 Objective Single cell of hippocampus was isolated, and mitochondria transmembrane potentials and the intracellular free calcium level [Ca2+]i were measured by means of flow cytometry (FCM) at the fifty-sixth day after mcmv infected.Methods Newborn mice of BALB/C were divided into two groups randomly; the virus inoculated group and the control group. After 56 days, the intracellular free calcium level and mitochondria transmembrane potentials in hippocampus were examined by FCM.Results The mitochondria transmembrane potentials was decreased (P<0.01) and the intracellular free calcium level [Ca2+]i was increased (P<0.01) in inoculated group.Conclusion The dysfunction of [Ca2+]i and mitochondria transmembrane potentials in hippocampus may play an important role in the CMV infected brain.
