黄芪注射液对高糖所致血管内皮细胞损伤的保护作用
Protective Effects of Huangqi on Vascular Endothelial Cells Exposed to High Gluose
【摘要】 背景与目的:研究黄芪注射液对50%葡萄糖所诱导的血管内皮细胞损伤的保护作用。材料与方法:采用血管内皮细胞VEC-304,分为正常对照组、葡萄糖损伤对照组和加入3个不同黄芪浓度的实验组(100mg/ml、200mg/ml、300mg/ml),各组细胞孵育24h后,除正常对照组外其余各组加入50%葡萄糖(终浓度为30mg/L)诱导损伤,继续培养4h。在倒置相差显微镜下观察细胞形态学变化并采用MTT光吸收法检测细胞活力、采用考马斯亮蓝法检测蛋白质含量,用RT-PCR法和免疫细胞化学方法检测血管内皮生长因子(VEGF mRNA)及蛋白的表达。结果:正常对照组及黄芪注射液各剂量组细胞活力、蛋白质含量均高于50%葡萄糖损伤对照组,其差异均具有统计学意义(P均<0.05)。显示高糖明显抑制VEGF的活性和表达,而黄芪则具有明显的对高糖损伤的修复作用。
【Abstract】 BACKGROUND AND AIM: To evaluate the protective effects of Huangqi on the damage of high glucose on vascular endothelial cell(VEC-304)in vitro.MATERIALS AND METHODS: The model of human umbilical vein endothelial cells(VEC-304) damage was induced by 50% glucose for 4 h.The cells were pretreated with three concentrations of Huangqi solution for 24 h,before they were exposed to glucose.MTT and protein levels in cells were determined after 4 h.The expression of VEGF was investigated by RT-PCR and immunohistochemistry methods.RESULTS: MTT and protein levels in cells of the normal control group amd Huangqi-treated groups were higher than in glucose-damaged group without huangqi(P<0.05).The expression of VEGF was inhibited by glucose.CONCLUSION: Huangqi could protect the damages of VEC-304 endothelial cells induced by high glucose.
