养阴活血方对转基因盐皮质激素受体过度表达小鼠模型心室重塑的影响
Effect of Nourishing Yin and Promoting Blood Circulation Formula on Ventricular Remodeling of Transgenic Mice with Mineralocorticoid Receptor Overexpression
【摘要】 目的观察养阴活血方对转基因(TG)盐皮质激素受体(MR)过度表达小鼠模型心室重塑的影响。方法将小鼠分为3组,即雄性非转基因小鼠组(NTG组)、雄性转基因小鼠组(TG组)和雄性转基因小鼠加中药组(TG+K组),心肌梗死造模后观察心脏重量和心脏重量/体重比、脑钠肽(BNP)的mRNA表达、生存率和心脏病理切片。结果养阴活血方干预治疗后,TG+K组心脏重量和心脏重量/体重比与TG组比较差异均有显著性意义(P<0.05),但与NTG组比较,差异无显著性意义(P>0.05);TG+K组的BNPmRNA较TG组表达减弱(P<0.05),较NTG组差异无显著性意义(P>0.05);TG+K组生存率高于TG组(P<0.05),与NTG组比较差异无显著性意义(P>0.05);心脏病理切片显示,TG组心脏纤维化明显,TG+K组心脏纤维化减轻,而NTG组最轻。结论养阴活血方可减轻雄性TGMR心肌梗死小鼠模型的心室重塑。
【Abstract】 Objective To observe the influence of Nourishing Yin and Promoting Blood Circulation Formula on ventricular remodeling of transgenic mice with mineralocorticoid receptor overexpression.Methods The mice were divided into three groups:male non-transgene(NTG)group,male transgene(TG)group,and male transgene + Chinese medicine(TG+herbs)group.After myocardial infarction model-making,the heart weight,ratio of heart weight to body weight,mRNA expression of brain natriuretic peptide,survival rate and heart pathological section were observed.Results With the herbal treatment to nourish yin and promote blood circulation,the heart weight and the ratio of heart weight to body weight of the TG+herbs group was significantly different from those of TG group(P<0.05),but the difference was not significant in comparison with NTG group(P>0.05).The mRNA expression of brain natriuretic peptide of TG+herbs group was weakened than that of TG group(P<0.05),but the difference was not significant in comparison with NTG group(P>0.05).The survival rate of TG+herbs group was higher than that of TG group(P<0.05),but the difference was not significant in comparison with NTG group(P>0.05).The heart pathological section showed an obvious fibrosis in TG group,a relieved fibrosis in TG+herbs group and a least fibrosis in NTG group.Conclusion The herbal treatment to nourish yin and promote blood circulation can relieve the ventricular remodeling of male TG mice with MR overexpression after myocardial infarction.
