西河柳中槲皮素与异鼠李素在植物体内分布及随季节变化
Variation and Distribution Quercetin and Isorhamnetin in the Cacumen Tamaricis by Season
【摘要】 目的:建立西河柳中槲皮素与异鼠李素的HPLC测定方法;研究槲皮素与异鼠李素在西河柳不同部位的分布及随季节变化的规律。方法:以NUCLEODUR100-5C18柱(150 mm×4.6 mm,5μm),甲醇-0.4%磷酸溶液(50:50)为流动相,流速为0.8ml·min-1,柱温为30℃,检测波长为360 nm。结果:槲皮素与异鼠李素线性范围分别为:0.17~1.01μg(r=0.999 7)和0.19~0.95μg(r=0.999 6),回收率分别为:102.3%,100.7%RSD4.0,3.1%;西河柳中槲皮素与异鼠李素含量随季节的变化而变化,且不同部位有不同的含量。结论:该方法可用于控制西河柳药材质量;西河柳中槲皮素与异鼠李素含量按春、夏、秋次序依次降低,在嫩技叶中含量最高。
【Abstract】 Objective:To establish a HPLC method for determination of quercetin and isorhamnetin in the Cacumen tamaricis and to study the variation and distribution laws of them in its parts in different seasons.Method:The Nucleodur100-5C18column(150 mm×4.6 mm,5μm)was used,the methanol-0.4% phosphoric acid solution(50:50)as mobile phase,the flow rate was 0.8 ml·min-1, the detection wavelength was 360 nm.Result:The linear ranges of quercetin and isorhomnetin were at 0.17 - 1.01μg(r=0.999 7) and 0.19-0.95(r=0.999 6)respectively.The average recoveries(n=6)were 102.3 % and 100.7 % with RSD 4.0% and 3.1% respectively.The content of quercetin and isorhamnetin in Cacumen tamaricis’s are variable by season and different in different parts. Conclusion:The method can be used for control the quality of Cacumen tamaricis.The content of quercetin and isorhamnetin in Cacu- men tamaricis are decrease in spring,summer and autumn sequence and the maximum is in tender-twig-leaves.
