系统性红斑狼疮患者Th1/Th2细胞因子的研究
Level and significance of Th1/Th2 cytokine in patients with systemic lupus erythe matosus
【摘要】 目的检测系统性红斑狼疮(SLE)患者外周白细胞介素-4(IL-4),白细胞介素-10(IL-10)、干扰素-γ(IFN-γ)和白细胞介素-2(IL-2)水平,探讨SLE患者的发病与Th1/Th2优势活化状态之间的关系。方法采用酶联免疫吸附法(ELISA)测定35例SLE患者和15名健康志愿者PBMC培养上清IL-4、IL-10、IL-2、IFN-γ水平。结果活动期SLE患者IL-4、IL-10水平明显高于非活动期、正常对照组,差异有显著性(P<0.01)。活动期SLE患者IL-2水平明显低于非活动期、正常对照组,差异有显著性(P<0.01)。IFN-γ水平3组间差异无显著性(P>0.05)。结论Th2型细胞因子介导的免疫应答在活动期SLE的发病机制中占主导地位。活动期SLE患者存在分泌型Th1细胞因子的细胞活化减弱。
【Abstract】 [Objective] To investigate interleukin-4 (IL-4), interleukin-10(IL-10), interferon gamma(ILN-γ)and interleukin-2 (IL-2) secreting capacity by peripheral blood mononuclear cells (PBMC) in 35 patients with systeimc lupus erythe matousus(SLE) and 15 healthy individuals, and to understand the role of Th1/Th2 balance in the pathogenesis of SLE. [Method] Sandwich ELISA was used to measure IL-4, IL-10,IFN-γ and IL-2 level in the supernatant of culture from PBMC. [Results] Compared with the static phase person and normal controls, the level of IL-4 and IL-10 produced spontaneously by PBMCs in active stage significantly increased (P <0.01). The level of IL-2 significantly decreased (P <0.01). The IFN-γ level was not statistically different between SLE patients and controls (P >0.05). [Conclusion] An imbalance towards Th2 predominace may play a significant role in the pathogenesis of SLE. Through investigating the levels of IL-4, IL-10and IL-2 of SLE, we offered the support to estimate the patients′ active states.
