PI3K抑制剂对转化生长因子β1诱导人肾小管上皮细胞Smad泛素化调节因子2表达的影响
Effect of LY294002 on expression of smad ubiquitin regulatory factor 2 induced by TGF-β1 in HK-2 cell line
【摘要】 目的探讨体外条件下转化生长因子β1(TGF-β1)对人近端肾小管上皮细胞系(HK-2)表达Smad泛素化调节因子2(Smurf2)的诱导情况,以及PI3K抑制剂LY294002对其的影响。方法应用RT-PCR法检测Smurf2 mRNA的表达,应用Western印迹法检测Smurf2蛋白的表达。结果HK-2细胞有基础量的Smurf2 mRNA和蛋白的表达,TGF-β1呈时间和剂量依赖性增加HK-2细胞Smurf2 mRNA和蛋白的表达(P<0.01)。PI3K抑制剂LY294002呈剂量依赖性抑制TGF-β1(100pM)所诱导的HK-2细胞Smurf2 mRNA和蛋白的表达(P<0.01)。结论在体外,TGF-β1可能通过PI3K/AKT通路诱导HK-2细胞Smurf2 mRNA和蛋白的表达。PI3K抑制剂LY294002可抑制Smurf2 mRNA和蛋白的表达。
【Abstract】 [Objective] To investigate the effect of inhibitor PI3K -LY294002 on the expression of Smurf 2 in cultured human proximal tubular epithelial cell line HK-2 induced by TGF-β1. [Methods] The mRNA expression was tested by RT-PCR. The protein expression was examined by Western blot. [Results] The basic levels of Smurf 2 mRNA and protein expression were observed in HK-2 cell line and up-regulated by TGF-β1 in dose-and time-dependent manner (P <0.01). The LY294002 significantly inhibited the enhanced Smurf 2 mRNA and protein expression in HK-2 cell line stimulated by TGF-β1 in dose-dependent manner (P <0.01). [Conclusion] TGF-β1 could up-regulate the expression of Smurf 2 mRNA and protein in vitro. The intracellular signaling transduction pathway is related to PI3K/Akt. LY294002 could inhibit the enhanced Smurf 2 mRNA and protein expression in HK-2 cell line stimulated by TGF-β1.
