【摘要】 目的观察CD25/IL-2Ra在人肝毛细胆管上的表达,以利于肝毛细胆管分布、形态的观察。方法引用免疫组化方法观察CD25/IL-2Ra分子在人肝毛细胆管上的表达,并设立阳性对照(扁桃体)和CD4、CD8之间的片间对照。通过免疫荧光法观察CD25/IL-2Ra在肝毛细胆管上的表达,结果与免疫组化相一致。通过阻断试验进一步确认肝毛细胆管上CD25/IL-2Ra分子表达的特异性。通过与国际公认的CEA(多克隆抗体)作对照,进一步确认CD25/IL-2Ra是一种优良的肝毛细胆管标记物。结果常规免疫组化方法DAB显色CD25/IL-2Ra标记的毛细胆管显棕黄色条索状结构,显示结果比国际公认的CEA更佳,免疫荧光呈索状绿色荧光表达,阻断试验进一步确认肝毛细胆管上CD25/IL-2Ra分子的存在。结论免疫组化、免疫荧光法检测CD25/IL-2Ra表达来观察肝脏毛细胆管的分布、形态等,具有明确的特异性。

【Abstract】 Objective To observe the expressions of CD25/IL-2Ra on cholangiole of human liver for observing the pathology of the cholangiole of human liver.Methods We observed the expressions of CD25/IL-2Ra on cholangiole of liver by immunohistochemisty stain,at the same time set up positive contrast(Tonsil tissue) and contrast of CD4,CD8.We observed the expressions of CD25/IL-2Ra on cholangiole of liver by immunofluorescence,results showed no difference.We can further confirm the specificity of the expressions of CD25/IL-2Ra on cholangiole of human liver by blocking test.We made a contrasting test of CEA(multi-clone immune body)which is generally acknowledged by the medical faculty and further confirm that CD25/IL-2Ra is a fine sign of cholangiole of human liver.Results The cholangiole signed by CD25/IL-2Ra showed yellowish-brown strip funicular structure and it’s effect is better than CEA’s.The expression of blue fluorescence is shown by immunofluorescence.The blocking test further confirms the existence of CD25/IL-2Ra on cholangiole of human liver.Conclusion Nowadays the pathologists make CEA the sign of cholangiole of human liver.We investigate the expressions of CD25/IL-2Ra by immunohistochemisty and immunofluorescence and observe the distribution and morphology of cholangiole of human liver.
