2006年杭州余杭区甲1亚型流感疫情的应急检测及基因特性分析
Rapid detection of character of influenza (H1N1) virus circulated in Yuhang in 2006
【摘要】 目的:对6起流感样疫情进行应急检测并对分离到的流行株的基因特征进行分析。方法:应用荧光RT-PCR的方法对6起流感样疫情进行检测并进行病毒分离,挑选H1N1亚型流感病毒的代表株进行血凝素基因(HA1)片断的核苷酸序列测定,分析基因和氨基酸变异情况。结果:应用荧光PCR检出甲1型流感阳性样本12例;与当前疫苗株A/Newcaledonia/20/1999相比,新分离到的流感株氨基酸序列有较大的差异,两者为不同性状的毒株。结论:H1N1亚型流感病毒重新引起流行并成为优势株,与其抗原性的变异及人群中针对这一病毒的免疫力下降有关。
【Abstract】 Objective:To detect and understand etiology character of influenza(H1N1) caused influenza outbreak of 2006 in Yuhang.Methods:We detected the influenza outbreak by real-time RT-PCR methods.Serological method was used to detect certain influenza virus′s type,representative influenza H1N1 virus was determined nucleotide sequence.Results:Twelve positive examples were detected by real-time RT-PCR.Compared to the vaccine A/New caledonia/20/1999,the sequence of HA1 that we isolate from the influenza outbreak occur obvious difference in amino acid sequence.Both were two different viruses.Conclusion:Influenza H1N1 virus preponderance may be because of the antigenicity variation and the crowd immunity as well as dropping of hemagglutinin(HA1) gene to analyses difference of the gene.
