应用玉米秸秆生产淡紫拟青霉36-1菌株孢子
Conidial Production of Paecilomyces lilacinus 36-1 with Cornstalk
【摘要】 为降低淡紫拟青霉36-1菌株生产成本,以玉米秸秆和玉米粉不同配比筛选,并通过添加氮源、碳源等单因子试验及玉米秸秆、玉米粉、米糠、硫酸铵等4个因子3水平正交试验。筛选出淡紫拟青霉36-1菌株固体发酵最佳产孢配方为:玉米秸秆∶米糠∶玉米粉=6∶4∶15(质量比),硫酸铵含量为1g/kg,含水量50%,其发酵产物产孢量为7.77×109个/g(干重)。发酵产物在室温下贮藏2~11个月,孢子萌发率仍达90%左右。
【Abstract】 Paecilomyces lilacinus 36-1,a nematophagous fungus,is of great potential for biocontrol of plant parasitic nematodes.The cornstalk was evaluated as a solid substrate for conidial production of P.lilacinus with lower cost.The results showed that the mycelial growth of P.lilacinus was obviously enhanced by addition of 0.1%-2% of urea and the conidial production was significantly increased by addition of 0.5% of ammonium sulfate,while no effect on growth and sporulation by supplement of 1%-4% of carbon sources.The orthogonal test L9(34) indicated that the optimum solid-state substrates components for the conidial production was cornstalk,rice bran and corn at a ratio of 6∶4∶15(weight),supplemented with 0.1% of ammonium sulfate and 50% of water.The conidial production was 7.77×109 conidia/g dry substrate after incubation at 25℃ for 10 d.The germination rate of P.lilacinus stored for 2-11 months at room temperature was about 90%.
