免疫球蛋白M在上皮源性恶性肿瘤及细胞系中的表达
Expression of Immunoglobulin M (IgM) in Multiple Epitheliogenic Malignant Tumor Tissue and Carcinoma Cell Lines
【摘要】 目的探讨IgM在多种上皮来源恶性肿瘤及肿瘤传代细胞系中的表达情况。方法应用组织芯片技术结合免疫组化SABC法检测IgM在10种上皮源性肿瘤及对应阴性组织中的表达情况,并通过流式细胞仪、间接免疫荧光法检测IgM在8种上皮源性肿瘤及Burkitt’s淋巴瘤(Raji)传代细胞系中的表达。结果IgM在10种上皮来源恶性肿瘤组织及部分对应阴性组织中均有表达,其阳性表达率分别为70.0%和20.0%(P<0.05);IgM在8种上皮来源恶性肿瘤传代细胞系及Raji细胞中存在表达,与阴性对照比较差异有统计学意义(P<0.05)。结论IgM在上皮来源恶性肿瘤及传代细胞系中存在广泛表达,其意义和机制有待进一步研究。
【Abstract】 Objective To examine the expression level of immunoglobulin M (IgM) in multiple epitheliogenic malignant tumor tissue and carcinoma cell lines. Methods Tissue microarray and immunohistochemistry were utilized to detect the expression level of IgM in ten epitheliogenic malignant tumor tissue and control negative tissue. Furthermore, flow cytometry and indirected immunofluorescence were utilized to detect the IgM expression level in eight epithelial carcinoma cell lines and the Burkitt’s lymphoma cell line (Raji). Results IgM was expressed in all ten epitheliogenic malignant tumor tissue. The overall positive expression rate of IgM in tumor tissue (70.0%) was significantly higher than that of control (20.0%) (P<0.05). IgM was also expressed in Raji and eight epithelial carcinoma cell lines. The positive IgM expression rate in each tumor cell line was significantly higher than that of control (P<0.05). Conclusion IgM is highly expressed in multiple epitheliogenic malignant tumor tissue and carcinoma cell lines when compared to negative control. Further study is necessary to fully understand the significance and mechanisms underlying the elevated IgM levels in epitheliogenic malignant tumors.
