茅苍术挥发油超临界萃取工艺及气质联用分析
Technology of extracting the essencial oil with supercritical from Atractylodes lancea(Thunb.)DC.and GC-MS analysis
【摘要】 目的:研究茅苍术挥发油超临界CO2萃取的最佳工艺条件和挥发油的化学成分。方法:以挥发油得率为指标,采用正交试验优选超临界CO2萃取茅苍术挥发油的最佳工艺;气质联用法(GC-MS)分析挥发油化学成分。结果:超临界CO2萃取茅苍术挥发油的最佳工艺为:萃取时间3.5 h,萃取温度45℃,萃取压力30 MPa,流量3 L/m in,挥发油得率为13.80%。GC-MS分析结果显示84个色谱峰,鉴定出67种化学成分,占总馏出峰面积的96.07%。含量最高的为苍术酮(24.92%),其次为苍术素(22.26%),β-桉叶醇(10.67%)等。结论:超临界CO2萃取茅苍术挥发油具有得率高,萃取时间短,温度低,无溶剂残留等优点。
【Abstract】 Objective:To study the optimal technical condition of extracting the essencial oil with supercritical CO2 from Atractylodes lancea(Thunb.)DC.and chemical composition in the oil.Methods:Optimized the technology of extracting the essencial oil with supercritical CO2 from Atractylodes lancea(Thunb.)DC.by simple factor and orthogonal experiment as guided by the yield of the essencial oil;GC-MS analyzed chemical composition. Results:The optimum technology of extracting the essencial oil with supercritical CO2 was as follows:3.5 h as extraction time,45℃as extraction temperature,30 MPa as extraction pressure,3 L/min as flow rate,13.80%as yield.The analytic result of GC-MS displayed 84 chromatographic peaks,identified 67 chemical compositions,that was 96.07 percent of whole run-down peak area.The supreme content was atractylone(24.92%),next atrctylodine(22.26%),β-eudesmol(10.67%)and so on. Conclusion:The SFE-CO2 was of advantage of:high yield,short extraction time,low temperature and without residue of solvent.
